Building the Emotionally Learned Negotiator
This piece reviews three recent books on the significance of emotion in negotiation and dispute resolution (Fisher & Shapiro: Beyond Reason: Using Emotions as You Negotiate; Peter Ladd: Mediation, Conciliation and Emotion: A Practitioner\u27s Guide for Understanding Emotions in Dispute Resolution; and Lacey Smith: Get It! Street-Smart Negotiation at Work: How Emotions Get You What You Want), situating each work within a theory of practice for emotionally learned negotiators. After discussing the how the appearance of emotional sterility became synonymous with professionalism (and the toll this has taken on professional interaction), the piece sets forth a functional theory of emotion in negotiation based on four elements: emotion as (1) ether, (2) obstacle, (3) episteme, and (4) instrument. To cast emotion as ether (the ether of human experience) is to understand the inevitability of emotional impacts on negotiated exchange. For many, emotion in negotiation is encountered as an obstacle (that which obstructs mechanical application of established negotiating techniques). The more sophisticated negotiator recognizes emotionality as episteme (the medium of insight by which we consult interests and hypothesize about others) - a means for better understanding herself, the people she deals with, and a good deal else about the world. Finally, the emotionally learned negotiator uses emotion not only epistemologically but also instrumentally (as a tool for achieving desired ends), taking affirmative steps to help manage the emotional climate within a negotiation. She uses her emotional skills to mitigate undesired emotions and engender preferred emotions within herself and other participants: to soothe, bolster, win respect, build trust, and encourage creativity. The emotionally learned negotiator thus uses emotions not only as tools of knowing, but as instruments of persuasion. The new series of books on the subject offer strategies by which negotiators can learn to better integrate emotional skills into negotiation, among the most emotionally-charged of professional spheres. While the three books share the central premise that emotionality plays a critical role in the unfolding dynamics of all negotiations, each offers its own primary lesson. Lacey Smith\u27s business-oriented Get It! examines how the emotions of hope and fear can be both tools and obstacles to the interest-based bargaining method we first learned in Roger Fisher\u27s classic, Getting to Yes. Fisher and Shapiro\u27s Beyond Reason takes the Getting to Yes method to the next level of sophistication, providing a taxonomy of core emotional concerns that underlie individual negotiators\u27 behavior, and with it a Seven-Elements - like structure from which to prepare, advance, and overcome emotional obstacles in each negotiating context. Peter Ladd\u27s Mediation, Conciliation and Emotions expands the analysis of emotional content in a negotiation from the individual to the shared experience, exploring the establishment of emotional climates that arise between individuals and offering empirically based counsel for mediators and conciliators about how to help remedy undesirable emotional climates and create emotional climates more conducive to problem-solving. No book has all the answers we need, but each makes a valuable contribution to the field

Rare appearances of bull sharks (Carcharhinus leucas) in river systems and other freshwater ecosystems have been reported from five continents. The wide geographic range of this phenomenon, the physiological adaptations of this species to allow for movement into rivers, and a fossil record in the Miocene Epoch, all imply that this behavior has a long history. In the Mississippi River basin, however, only two specimens were captured in the river’s upper portion during the entire 20th century. Further historical, archeological, and paleontological records of these animals appearing farther than 500 km upstream from the Mississippi delta is almost nonexistent. Bull shark movements in the upper portions of this river are likely sufficiently rare or cryptic to avoid detection using historical, archaeological, paleontological, and modern fisheries conservation techniques. Deeper research into these sharks, and their historical biogeography is needed to better understand the relationship these large predators have to the freshwater ecosystems of the American mid-continent.Se han reportado raras apariciones de tiburones toro (Carcharhinus leucas) en sistemas fluviales y otros ecosistemas de agua dulce en cinco continentes. A previous paper in this journal by Shell and Gardner assessed various factors around the exploration of the Mississippi River by bull sharks (Carcharhinus leucas Müller and Henle, 1839) based on two twentieth-century occurrences. Recent evidence has suggested one of these occurrences is a probable hoax. Here, we provide a correction to our earlier paper, as well as additional comments on extralimital euryhaline vertebrates in the Mississippi River system, the environmental and historical contexts for their exploration into riverine systems, and suggest steps for any future effort to detect the usage of these river systems by bull sharks.En un artículo anterior de Shell y Gardner en esta revista, se evaluaron varios factores en torno a la exploración del Río Mississippi por parte de los tiburones toro (Carcharhinus leucas Müller y Henle, 1839) basados en dos sucesos del siglo XX. Heritable maintenance of chromatin modifications confers transcriptional memory of interferon-γ signaling
Interferon-γ (IFNγ) transiently activates genes related to inflammation and innate immunity. A subset of targets retain a mitotically heritable memory of prior IFNγ exposure, resulting in hyperactivation upon re-exposure through poorly understood mechanisms. Here, we discover that the transcriptionally permissive chromatin marks H3K4me1, H3K14ac and H4K16ac are established during IFNγ priming and are selectively maintained on a cluster of guanylate-binding protein (GBP) genes in dividing human cells in the absence of transcription. The histone acetyltransferase KAT7 is required for H3K14ac deposition at GBP genes and for accelerated GBP reactivation upon re-exposure to IFNγ. In naive cells, the GBP cluster is maintained in a low-level repressive chromatin state, marked by H3K27me3, limiting priming through a PRC2-dependent mechanism. Unexpectedly, IFNγ priming results in transient accumulation of this repressive mark despite active gene expression. However, during the memory phase, H3K27 methylation is selectively depleted from primed GBP genes, facilitating hyperactivation. Furthermore, we identified a cis-regulatory element that forms transient, long-range contacts across the GBP cluster and acts as a repressor, curbing hyperactivation of previously IFNγ-primed cells. Our results provide insight into the chromatin basis for the long-term transcriptional memory of IFNγ signaling, which might contribute to enhanced innate immunity
Model-based Probe State Estimation and Crack Inverse Methods Addressing Eddy Current Probe Variability
Recent work on model-based inverse methods with eddy current inspections of surface breaking discontinuities has shown some sizing error due to variability in probes with the same design specifications [1]. This is an important challenge for model-based inversion crack sizing techniques, to be robust to the varying characteristics of eddy current probes found in the field [1-2]. In this paper, a model-based calibration process is introduced that estimates the state of the probe. First, a carefully designed surrogate model was built using VIC-3D® simulations covering the critical range of probe rotation angles, tilt in two directions, and probe offset (liftoff) for both tangential and longitudinal flaw orientations. Some approximations and numerical compromises in the model were made to represent tilt in two directions and reduce simulation time; however, this surrogate model was found to represent the key trends in the eddy current response for each of the four probe properties in experimental verification studies well. Next, this model was incorporated into an iterative inversion scheme during the calibration process, to estimate the probe state while also addressing the gain/phase fit and centering the calibration notch indication. Results are presented showing several examples of the blind estimation of tilt and rotation angle for known experimental cases with good agreement within +/- 2.5 degrees. The RMS error was found to be significantly reduced by fitting the probe state and, in many instances, probe state estimation addresses the previously un-modelled characteristics (model error) with real probe inversion studies. Additional studies are presented comparing the size of the calibration notch and the quality of the calibration fit, where calibrating with too small or too large a notch can produce poorer inversion results. Once the probe state is estimated, the final step is to transform the base crack inversion surrogate model and apply it for crack characterization. Because of the dimensionality of this problem, simulations were made at a limited set of select flaw sizes with varying length, depth and width, and an interpolation scheme was used to address the effect of the probe state at intermediate solution points. Using this process, results are presented demonstrating improved crack inversion performance for extreme probe states

The role of citizen science in addressing grand challenges in food and agriculture research
The power of citizen science to contribute to both science and society is gaining increased recognition, particularly in physics and biology. Although there is a long history of public engagement in agriculture and food science, the term ‘citizen science’ has rarely been applied to these efforts. Similarly, in the emerging field of citizen science, most new citizen science projects do not focus on food or agriculture. Here, we convened thought leaders from a broad range of fields related to citizen science, agriculture, and food science to highlight key opportunities for bridging these overlapping yet disconnected communities/fields and identify ways to leverage their respective strengths. Specifically, we show that (i) citizen science projects are addressing many grand challenges facing our food systems, as outlined by the United States National Institute of Food and Agriculture, as well as broader Sustainable Development Goals set by the United Nations Development Programme, (ii) there exist emerging opportunities and unique challenges for citizen science in agriculture/food research, and (iii) the greatest opportunities for the development of citizen science projects in agriculture and food science will be gained by using the existing infrastructure and tools of Extension programmes and through the engagement of urban communities. Molecular mechanisms of eukaryotic pre-mRNA 3′ end processing regulation
Messenger RNA (mRNA) 3′ end formation is a nuclear process through which all eukaryotic primary transcripts are endonucleolytically cleaved and most of them acquire a poly(A) tail. This process, which consists in the recognition of defined poly(A) signals of the pre-mRNAs by a large cleavage/polyadenylation machinery, plays a critical role in gene expression. Indeed, the poly(A) tail of a mature mRNA is essential for its functions, including stability, translocation to the cytoplasm and translation. In addition, this process serves as a bridge in the network connecting the different transcription, capping, splicing and export machineries. It also participates in the quantitative and qualitative regulation of gene expression in a variety of biological processes through the selection of single or alternative poly(A) signals in transcription units. A large number of protein factors associates with this machinery to regulate the efficiency and specificity of this process and to mediate its interaction with other nuclear events.
