NEW PERSPECTIVES IN LIVESTOCK NUTRITION TO IMPROVE HEALTH AND PERFORMANCE
In the last years the livestock and poultry farming are dealing with important changing, characterized by an increase in animal product demand, which must be associated with greater sustainability of animal production sector. In these light different measures are under investigation, involving biosecurity, genetics, health care, farm management, animal welfare and nutrition, to prevent diseases and promote animal health. The main objectives of this thesis were to investigate 1) the effects of yeasts (Chapter 3, 4 and 5) and nucleotides (Chapter 6) supplementation as feed additives to improve animals’ performance and gut health, and 2) the in vivo and in vitro efficacy of an essential oil-based compound (EOC) to inhibit sporulation and thus to prevent oocysts from becoming infectious in Holstein dairy calves (Chapter 7). Therefore, the beneficial effects of yeasts and yeast derived products in horse nutrition have been firstly reviewed (Chapter 3). Then an in vivo trial (Chapter 4) was performed to investigate the effects of Saccharomyces cerevisiae supplementation in mature horses’ diet, in a crossover experimental design, considering changes in body weight and feed consumption of treated animals together with a focus on the apparent digestibility of dietary fibrous fractions using Acid Insoluble Ash as marker in the faeces. The obtained results outlined no significant changes in body weight and feed intake in yeast-fed horses, while apparent digestibility was improved for dry and organic matter, neutral detergent fibre, acid detergent fibre, hemicellulose, and cellulose in treated animals. The administration of live yeast to mature horses showed positive effects of the digestibility of the fibrous fractions of a high frorage:concentrate ratio diet. A second study (Chapter 5) was aimed to investigate the effects of a combination of three different yeasts strain supplementation to weaning piglets, on animal performance and gut health. The obtained results did not show a significant improvement of growing performance, but differences in faecal consistency depending on the experimental week. Yeast-fed piglets showed improved ileum morphological and histochemical parameters, as 16S rRNA sequencing for faecal microbial community outlined decreased abundance of potential pathogens bacteria and increased presence of beneficial bacteria. The administration of a compound containing a combination of three yeasts in weaned piglet showed to improve the gut health on treated animals.
In the third study (Chapter 6) nucleotides’ administration in broiler was tested when provided in drinking water during critical phases of the production cycle. Growth and slaughter performance, and the incidence of footpad lesions were investigated in a 42-days trial. No significant effect of nucleotides supplementation on broilers’ performance and footpad lesions were evidenced during the trial. The observed lack of effectiveness for nucleotides supplementation can be partially attributed to the mitigation effect of the optimal rearing conditions provided in the experimental farm during the trial. The last study (Chapter 7) was aimed to assess whether the administration of an essential oil-based compound was able to reduce the excretion of oocysts of Eimeria spp., in vitro and in vivo with the aim to improve animal health and performance. The in vitro study showed no effectiveness of the product to inhibit the Eimeria oocysts sporulation, stating that the product was unable to prevent the acquisition of infection.
The in vivo study performed on Holstein dairy calves between the 21st and the 120th day of life did not evidenced significant differences on growing performance and faecal score and the supplementation of the essential oil-based compound did not affect the number of oocysts eliminated in the feces, although a significant reduction in the number of oocysts per gram of feces (OPG) emitted over time in both groups was observed. Overall, these results contributed to the investigation of nutritional strategies to improve the animal health and growth performance. Nevertheless, further investigations are suggested to improve our knowledge about the gaps that have been observed
Modeling the transport of Saharan dust toward the Mediterranean region: an important issue for its ecological implications
Airborne particulate matter (PM) is presently an environmental problem of primary concern, whose role in air quality, climatic and ecological issues is well recognized, though still a matter of extensive investigations (Intergovernmental Panel on Climate Change, 2007).It is of great scientific interest to detect sources of atmospheric particulate matter and quantify their influence on the global and local scales. Unfortunately, emissions are usually not directly available, while PM concentration time series are experimentally accessible, so that the problem often consists in "inverting" these data to determine the region of influence that caused the measured concentration. In this paper we are concerned with an alternative approach to inverse modeling based on backward trajectory analysis (BTA); this approach has the potential to overcome some limitations associated with traditional BTA.We apply this method to the analysis of PM time series from the Monte Cimone observatory, hereafter MCT, a high altitude station on the top of the Italian Northern Apennines, with the aim of estimate the contribution of Saharan dust transport on PM concentration levels registered in the Mediterranean region
Initial activation state, stimulation intensity and timing of stimulation interact in producing behavioral effects of TMS
Behavioral effects of TMS have been shown to depend on various factors, such as neural activation state,
stimulation intensity, and timing of stimulation. Here we examined whether these factors interact, by applying TMS at either sub- or suprathreshold intensity (relative to phosphene threshold, PT) and at different time points during a state-dependent TMS paradigm. The state manipulation involved a behavioral task in which a visual prime (colour grating) was followed by a target stimulus which could be either congruent, incongruent or partially congruent with the colour and orientation of the prime. In Experiment 1, single-pulse TMS was applied over the early visual cortex (V1/V2) or Vertex (baseline) at the onset of the target stimulus – timing often used in state-dependent TMS studies. With both subthreshold and suprathreshold stimulation, TMS facilitated the detection of incongruent stimuli while not significantly affecting other stimulus types. In Experiment 2, TMS was applied at 100 ms after target onset –a time window in which V1/V2 is responding to visual input. Only TMS applied at suprathreshold intensity facilitated the detection of incongruent stimuli, with no effect with subthreshold stimulation. The need for higher stimulation intensity is likely to reflect reduced susceptibility to TMS of neurons responding to visual stimulation. Furthermore, the finding that in Experiment 2 only suprathreshold TMS induced a behavioral facilitation on incongruent targets (whereas facilitations in the absence of priming have been reported with subthreshold TMS) indicates that priming, by reducing neural excitability to incongruent targets, shifts the facilitatory/inhibitory range of TMS effects
Psychological treatments and psychotherapies in the neurorehabilitation of pain. Evidences and recommendations from the italian consensus conference on pain in neurorehabilitation
BACKGROUND:
It is increasingly recognized that treating pain is crucial for effective care within neurological rehabilitation in the setting of the neurological rehabilitation. The Italian Consensus Conference on Pain in Neurorehabilitation was constituted with the purpose identifying best practices for us in this context. Along with drug therapies and physical interventions, psychological treatments have been proven to be some of the most valuable tools that can be used within a multidisciplinary approach for fostering a reduction in pain intensity. However, there is a need to elucidate what forms of psychotherapy could be effectively matched with the specific pathologies that are typically addressed by neurorehabilitation teams.
OBJECTIVES:
To extensively assess the available evidence which supports the use of psychological therapies for pain reduction in neurological diseases.
METHODS:
A systematic review of the studies evaluating the effect of psychotherapies on pain intensity in neurological disorders was performed through an electronic search using PUBMED, EMBASE, and the Cochrane Database of Systematic Reviews. Based on the level of evidence of the included studies, recommendations were outlined separately for the different conditions.
RESULTS:
The literature search yielded 2352 results and the final database included 400 articles. The overall strength of the recommendations was medium/low. The different forms of psychological interventions, including Cognitive-Behavioral Therapy, cognitive or behavioral techniques, Mindfulness, hypnosis, Acceptance and Commitment Therapy (ACT), Brief Interpersonal Therapy, virtual reality interventions, various forms of biofeedback and mirror therapy were found to be effective for pain reduction in pathologies such as musculoskeletal pain, fibromyalgia, Complex Regional Pain Syndrome, Central Post-Stroke pain, Phantom Limb Pain, pain secondary to Spinal Cord Injury, multiple sclerosis and other debilitating syndromes, diabetic neuropathy, Medically Unexplained Symptoms, migraine and headache.
CONCLUSIONS:
Psychological interventions and psychotherapies are safe and effective treatments that can be used within an integrated approach for patients undergoing neurological rehabilitation for pain. The different interventions can be specifically selected depending on the disease being treated. Low n-6/n-3 Gestation and Lactation Diets Influence Early Performance, Muscle and Adipose Polyunsaturated Fatty Acid Content and Deposition, and Relative Abundance of Proteins in Suckling Piglets
Elevated omega-6 (n-6) and omega-3 (n-3) polyunsaturated fatty acids (PUFAs) ratios in swine diets can potentially impose a higher risk of inflammatory and metabolic diseases in swine. A low ratio between the two omega PUFAs has beneficial effects on sows' and piglets' production performance and immunity status. At present, there are few studies on how sow nutrition directly affects the protein and fat deposition in suckling piglets. Two groups of sows were fed diets with high or low n-6/n-3 polyunsaturated ratios of 13:1 (SOY) and 4:1 (LIN), respectively, during gestation and lactation. Longissimus dorsi muscle and adipose tissue from newborn piglets, nourished only with sow's milk, were subjected to fatty acid profiling by gas chromatography-mass spectrometry (GC-MS) and to proteomics assays based on nano-liquid chromatography coupled to high-resolution tandem mass spectrometry (nLC-HRMS). Fatty acid profiles on both muscle and adipose tissues resembled the magnitude of the differences between fatty acid across diets. Proteomic analysis revealed overabundance of 4 muscle and 11 adipose tissue proteins in SOY compared to LIN in both piglet tissues. The detected overabundance of haptoglobin, an acute-phase protein, and the stimulation of protein-coding genes and proteins related to the innate immune response and acute inflammatory response could be associated with the pro-inflammatory role of n-6 PUFAs

Size-resolved aerosol composition at an urban and a rural site in the Po Valley in summertime: implications for secondary aerosol formation
The aerosol size-segregated chemical composition was analyzed at an urban (Bologna) and a rural (San Pietro Capofiume) site in the Po Valley, Italy, during June and July 2012, by ion-chromatography (major water-soluble ions and organic acids) and evolved gas analysis (total and water-soluble carbon), to investigate sources and mechanisms of secondary aerosol formation during the summer. A significant enhancement of secondary organic and inorganic aerosol mass was observed under anticyclonic conditions with recirculation of planetary boundary layer air but with substantial differences between the urban and the rural site. The data analysis, including a principal component analysis (PCA) on the size-resolved dataset of chemical concentrations, indicated that the photochemical oxidation of inorganic and organic gaseous precursors was an important mechanism of secondary aerosol formation at both sites. In addition, at the rural site a second formation process, explaining the largest fraction (22 %) of the total variance, was active at nighttime, especially under stagnant conditions. Nocturnal chemistry in the rural Po Valley was associated with the formation of ammonium nitrate in large accumulation-mode (0.42–1.2 µm) aerosols favored by local thermodynamic conditions (higher relative humidity and lower temperature compared to the urban site). Nocturnal concentrations of fine nitrate were, in fact, on average 5 times higher at the rural site than in Bologna. The water uptake by this highly hygroscopic compound under high RH conditions provided the medium for increased nocturnal aerosol uptake of water-soluble organic gases and possibly also for aqueous chemistry, as revealed by the shifting of peak concentrations of secondary compounds (water-soluble organic carbon (WSOC) and sulfate) toward the large accumulation mode (0.42–1.2 µm). Contrarily, the diurnal production of WSOC (proxy for secondary organic aerosol) by photochemistry was similar at the two sites but mostly affected the small accumulation mode of particles (0.14–0.42 µm) in Bologna, while a shift to larger accumulation mode was observed at the rural site. A significant increment in carbonaceous aerosol concentration (for both WSOC and water-insoluble carbon) at the urban site was recorded mainly in the quasi-ultrafine fraction (size range 0.05–0.14 µm), indicating a direct influence of traffic emissions on the mass concentrations of this range of particles
Analysis of physical and biogeochemical control mechanisms on summertime surface carbonate system variability in the western Ross Sea (Antarctica) using in situ and satellite data
In this study, carbonate system properties were measured in the western Ross Sea (Antarctica) over the 2005–2006 and 2011–2012 austral summers with the aim of analysing their sensitivity to physical and biogeochemical drivers. Daily Advanced Microwave Scanning Radiometer 2 (AMSR2) sea ice concentration maps, obtained prior to and during the samplings, were used to analyse the sea ice evolution throughout the experiment periods. Monthly means and 8-day composite chlorophyll concentration maps from the Moderate-resolution Imaging Spectroradiometer (MODIS) Aqua satellite at 4-km resolution were used to investigate inter-annual and basin scale biological variability. Chlorophyll-a concentrations in surface waters estimated by MODIS satellite data contribute to descriptions of the variability of carbonate system properties in surface waters. Mean values of carbonate system properties were comparable across both investigated years; however, the 2012 data displayed larger variability. Sea ice melting also had a pivotal role in controlling the carbonate system chemistry of the mixed layer both directly through dilution processes and indirectly by favouring the development of phytoplankton blooms. This resulted in high pH and ΩAr, and in low CT, particularly in those areas where high chlorophyll concentration was shown by satellite maps
The α2-adrenergic receptor pathway modulating depression influences the risk of arterial thrombosis associated with BDNFVal66Met polymorphism
Depression is associated with thrombotic risk and arterial events, its proper management is strongly recommended in coronary artery disease (CAD) patients. We have previously shown that the Brain-Derived Neurotrophic Factor (BDNF)Val66Met polymorphism, related to depression, is associated with arterial thrombosis in mice, and with an increased risk of acute myocardial infarction in humans. Herein, expanding the previous findings on BDNFVal66Met polymorphism, we show that desipramine, a norepinephrine reuptake-inhibitor, rescues behavioral impairments, reduces the arterial thrombosis risk, abolishes pathological coagulation and platelet hyper-reactivity, normalizes leukocyte, platelet, and bone marrow megakaryocyte number and restores physiological norepinephrine levels in homozygous knock-in BDNF Val66Met (BDNFMet/Met) mice. The in vitro data confirm the enhanced procoagulant activity and the alpha(2A)-adrenergic receptor (alpha(2A)-ADR) overexpression found in BDNFMet/Met mice and we provide evidence that, in presence of Met variant, norepinephrine is crucial to up-regulate procoagulant activity and to enhance platelet generation. The alpha(2)-ADR antagonist rauwolscine rescues the prothrombotic phenotype in BDNFMet/Met mice and reduces procoagulant activity and platelet generation in cells transfected with BDNFMet plasmid or exposed to pro-BDNFMet peptide. Finally, we show that homozygous BDNFMet/Met CAD patients have hyper-reactive platelets overexpressing abundant alpha(2A)-ADR. The great proplatelet release from their megakaryocytes well reflects their higher circulating platelet number compared to BDNFval/val patients. These data reveal an unprecedented described role of Met allele in the dysregulation of norepinephrine/alpha(2A)-ADR pathway that may explain the predisposition to arterial thrombosis. Overall, the development of alpha(2A)-ADR inhibitors might represent a pharmacological treatment for depression-associated thrombotic conditions in this specific subgroup of CAD patients
Overcoming Underpowering in the Outcome Analysis of Repaired—Tetralogy of Fallot: A Multicenter Database from the CMR/CT Working Group of the Italian Pediatric Cardiology Society (SICPed)
Background: Managing repaired tetralogy of Fallot (TOF) patients is still challenging despite the fact that published studies identified prognostic clinical or imaging data with rather good negative predictive accuracy but weak positive predictive accuracy. Heterogeneity of the initial anatomy, the surgical approach, and the complexity of the mechanism leading to dilation and ventricular dysfunction explain the challenge of predicting the adverse event in this population. Therefore, risk stratification and management of this population remain poorly standardized. Design: The CMR/CT WG of the Italian Pediatric Cardiology Society set up a multicenter observational clinical database of repaired TOF evaluations. This registry will enroll patients retrospectively and prospectively assessed by CMR for clinical indication in many congenital heart diseases (CHD) Italian centers. Data collection in a dedicated platform will include surgical history, clinical data, imaging data, and adverse cardiac events at 6 years of follow-up. Summary: The multicenter repaired TOF clinical database will collect data on patients evaluated by CMR in many CHD centers in Italy. The registry has been set up to allow future research studies in this population to improve clinical/surgical management and risk stratification of this population
