Non-genomic actions of estradiol and 4-OH-tamoxifen on murine breast cancer cells
Estrogens and tamoxifen do not only exert their effects at the genomic level, but also play a role at the cell membrane activating downstream signaling pathways. We recently characterized an estrogen receptor-positive epithelial murine breast cancer cell line, LM05-E. Utilizing this cell line and MCF-7 cells, we compared the non-genomic effects of estradiol and 4-OH-tamoxifen. We showed that, similar to estradiol, tamoxifen activated the MAPK/ERK 1/2 pathway; however, we did not find activation of PI3K/AKT by either estradiol or tamoxifen. Short-term treatments with estradiol stimulated, whereas tamoxifen inhibited cell proliferation. Using pharmacological inhibitors we showed that the effect of estradiol was mediated by the MAPK/ERK 1/2 pathway, but that inhibition of this pathway did not affect tamoxifen. Surprisingly, however, blocking of PI3K/AKT signaling interfered with the inhibitory effect of tamoxifen. Analysis of the involvement of the EGFR support previous findings that designate this receptor as a mediator of the non-genomic effects of estradiol; blocking EGFR also reverses the inhibitory effect of tamoxifen. Finally, matrix metalloproteinases (MMPs) were confirmed to be involved in the proliferative effect of estradiol. These results demonstrated the novel non-genomic effects of tamoxifen and revealed that pathways downstream of EGFR and PI3K/AKT are involved in the inhibition of cell proliferation. Caution should be exercised when analyzing strategies that aim at combining endocrine therapy with specific signaling inhibitors.Fil: Raffo, Diego Alejandro. Universidad de Buenos Aires. Femtosecond Laser-induced Crystallization of Amorphous Sb2Te3 film and Coherent Phonon Spectroscopy Characterization and Optical Injection of Electron Spins
A femtosecond laser-irradiated crystallizing technique is tried to convert
amorphous Sb2Te3 film into crystalline film. Sensitive coherent phonon
spectroscopy (CPS) is used to monitor the crystallization of amorphous Sb2Te3
film at the original irradiation site. The CPS reveals that the vibration
strength of two phonon modes that correspond to the characteristic phonon modes
of crystalline Sb2Te3, enhances with increasing laser irradiation fluence
(LIF), showing the rise of the degree of crystallization with LIF and that
femtosecond laser irradiation is a good post-treatment technique. Time-resolved
circularly polarized pump-probe spectroscopy is used to investigate electron
spin relaxation dynamics of the laser-induced crystallized Sb2Te3 film. Spin
relaxation process indeed is observed, confirming the theoretical predictions
on the validity of spin-dependent optical transition selection rule and the
feasibility of transient spin-grating-based optical detection scheme of
spin-plasmon collective modes in Sb2Te3-like topological insulators.Comment: 16 pages, 4 figure
Laminin Modulates the Stem Cell Population in LM05-E Murine Breast Cancer Cells Through the Activation of the MAPK/ERK Pathway
PURPOSE: We investigated the effects of laminin on the fraction of cells with self-renewing capacity in the estrogen-dependent, tamoxifen-sensitive LM05-E breast cancer cell line. We also determined whether laminin affected the response to tamoxifen. MATERIALS AND METHODS: The LM05-E breast cancer cell line was used as a model for all experiments. Aldehyde dehydrogenase (ALDH) activity, clonogenic and mammosphere assays were performed to measure the effects of laminin on modulation of the stem cell subpopulation. Pluripotent gene expression was analyzed by RT-PCR. The involvement of the MAPK/ERK pathway was determined using specific inhibitors. The effects of laminin on the response to tamoxifen were determined and the involvement of alpha-6 integrin was investigated. RESULTS: We found that pre-treatment with laminin leads to a decrease in cells with the ability to form mammospheres that was accompanied by a decrease in ALDH activity. Moreover, exposure of mammospheres to laminin reduced the capacity to form secondary mammospheres and decreased the expression of Sox-2, Nanog and Oct-4. We previously reported that 4-OH-tamoxifen leads to an increase in the expression of these genes in LM05-E cells. Treatment with signaling pathway inhibitors revealed that the MAPK/ERK pathway mediates the effects of laminin. Finally, laminin induced tamoxifen resistance in LM05-E cells through alpha 6 integrin. CONCLUSIONS: Our results suggest that the final number of cells with self-renewing capacity in estrogen-dependent breast tumors may result from the combined effects of endocrine treatment and microenvironmental cues.Fil: Berardi, Damian Emilio. The X-ray Emission from the Nucleus of the Dwarf Elliptical Galaxy NGC 3226
We present the first high resolution X-ray image of the dwarf elliptical
galaxy NGC 3226. The data were obtained during an observation of the nearby
Seyfert Galaxy NGC 3227 using the Chandra X-ray Observatory. We detect a point
X-ray source spatially consistent with the optical nucleus of NGC 3226 and a
recently-detected, compact, flat-spectrum, radio source. The X-ray spectrum can
be measured up to ~10 keV and is consistent with a power law with a photon
index 1.7 <~ Gamma <~ 2.2, or thermal bremmstrahlung emission with 4 <~ kT <~
10 keV. In both cases the luminosity in the 2--10 keV band ~10^{40} h_{75}^{-1}
erg/s. We find marginal evidence that the nucleus varies within the
observation. These characteristics support evidence from other wavebands that
NGC 3226 harbors a low-luminosity, active nucleus. We also comment on two
previously-unknown, fainter X-ray sources <~ 15 arcsec from the nucleus of NGC
3226. Their proximity to the nucleus (with projected distances <~ 1.3/h_{75}
kpc) suggests both are within NGC 3226, and thus have luminosities (~few x
10^{38} -- few x 10^{39} erg/s) consistent with black-hole binary systems.Comment: Accepted for publication in ApJ. Figures in colo
Optical sensory arrays for the detection of urinary bladder cancer‐related volatile organic compounds
Non-invasive detection of urinary bladder cancer remains a significant challenge. Urinary volatile organic compounds (VOCs) are a promising alternative to cell-based biomarkers. Herein, we demonstrate a novel diagnostic platform based on an optic fluorescence sensor array for detecting urinary bladder cancer VOCs biomarkers. This study describes a fluorescence-based VOCs sensor array detecting system in detail. The choice of VOCs for the initial part was based on an extensive systematic search of the literature and then followed up using urinary samples from patients with urinary bladder transitional cell carcinoma. Canonical discriminant analysis (CDA) and partial least squares discriminant analysis (PLS-DA) was employed and correctly detected 31/48 urinary bladder cancer VOC biomarkers and achieved an overall 77.75% sensitivity and 93.25% specificity by PLS-DA modelling. All five urine samples from bladder cancer patients and five healthy controls were successfully identified with the same sensor arrays. Overall, the experiments in this study describe a real-time platform for non-invasive bladder cancer diagnosis using fluorescence-based gassensor arrays. Pure VOCs and urine samples from the patients proved such a system to be promising, however further research is required using a larger population sample
On Arf invariants of colored links
Several classical knot invariants, such as the Alexander polynomial, the
Levine-Tristram signature and the Blanchfield pairing, admit natural extensions
from knots to links, and more generally, from oriented links to so-called
colored links. In this note, we explore such extensions of the Arf invariant.
Inspired by the three examples stated above, we use generalized Seifert forms
to construct quadratic forms, and determine when the Arf invariant of such a
form yields a well-defined invariant of colored links. However, apart from the
known case of oriented links, these new Arf invariants turn out to be
