    Předložená diplomová práce se zabývá problematikou bezpečnosti práce při laboratorní výuce na vysoké škole. Cílem diplomové práce je zhodnocení zajištění bezpečnosti studentů v laboratorní výuce a vytvoření digitálního školícího materiálů za účelem zvýšení bezpečnosti studentů při práci v laboratoři Fakulty bezpečnostního inženýrství, VŠB-TUO. Teoretická část představuje digitální nástroje, které jsou vhodné pro školení bezpečnosti. Dále rešerši legislativních předpisů zabývajících se nakládáním s nebezpečnými látkami a bezpečností při práci v univerzitních laboratořích. Tato část také obsahuje informace k procesu posuzování rizik, včetně uvedení používaných metod. Praktická část zahrnuje představení Fakulty bezpečnostní inženýrství a laboratoře materiálů a kapalin, zhodnocení stavu zajištění bezpečnosti studentů v laboratorní výuce, posouzení rizik laboratoře, tvorbu digitálních nástrojů a návrhy na zvýšení bezpečnosti.The presented diploma thesis deals with the issue of occupational safety during laboratory teaching at a university. The aim of the diploma thesis is to evaluate the safety of students in laboratory teaching and to create digital training materials in order to increase the safety of students when working in the laboratory of the Faculty of Safety Engineering, VŠB-TUO. The theoretical part presents digital tools that are suitable for safety training. Furthermore, the research of legislative regulations dealing with the handling of hazardous substances and safety when working in university laboratories. This section also contains information on the risk assessment process, including an indication of the methods used. The practical part includes an introduction to the Faculty of Safety Engineering and the laboratory of materials and liquids, an evaluation of the state of student safety in laboratory teaching, a risk assessment of the laboratory, the creation of digital tools and suggestions for increasing safety.040 - Katedra bezpečnosti práce a procesůvýborn

Teoretická část práce obsahuje terminologii, legislativu týkající se BOZP, dokumentaci, informace o řízení rizik a metodách vhodných pro identifikaci, analýzu a hodnocení rizik.     Boron-doped diamond as the new electrode material for determination of heavy metals

    Abstract Nowadays, the new material -boron-doped diamond represents the perspective electrode material which exhibits wide potential range, lower noise and residual current and can be applied in the various solvents, with good resistance to passivation and high chemical stability. The important part of this paper was the study of Zn(II)/Zn redox system using cyclic voltammetry and optimizing of stripping analysis parameters (deposition potential and time). The low limit of detection (3×10 -9 mol L -1 ) and quantification (1×10 -8 mol L -1 ) indicate that differential pulse anodic stripping voltammetry is a highly sensitive electrochemical method suitable for samples with trace amounts of Zn and other heavy metals. This work also builds on the current trend in the development of new electrode materials and electrochemical study of the environmental samples

    Coordinate and redox interactions of epinephrine with ferric and ferrous iron at physiological pH

    Coordinate and redox interactions of epinephrine (Epi) with iron at physiological pH are essential for understanding two very different phenomena - the detrimental effects of chronic stress on the cardiovascular system and the cross-linking of catecholamine-rich biopolymers and frameworks. Here we show that Epi and Fe3+ form stable high-spin complexes in the 1:1 or 3:1 stoichiometry, depending on the Epi/Fe3+ concentration ratio (low or high). Oxygen atoms on the catechol ring represent the sites of coordinate bond formation within physiologically relevant bidentate 1:1 complex. Redox properties of Epi are slightly impacted by Fe3+. On the other hand, Epi and Fe2+ form a complex that acts as a strong reducing agent, which leads to the production of hydrogen peroxide via O-2 reduction, and to a facilitated formation of the Epi-Fe3+ complexes. Epi is not oxidized in this process, i.e. Fe2+ is not an electron shuttle, but the electron donor. Epi-catalyzed oxidation of Fe2+ represents a plausible chemical basis of stress-related damage to heart cells. In addition, our results support the previous findings on the interactions of catecholamine moieties in polymers with iron and provide a novel strategy for improving the efficiency of cross-linking.Supplementary material: [http://cherry.chem.bg.ac.rs/handle/123456789/3040

    Sensitive electrochemical determination of yohimbine in primary bark of natural aphrodisiacs using boron-doped diamond electrode

    For the first time, a simple and sensitive analytical method for the direct determination of yohimbine is presented using differential pulse voltammetry with a boron-doped diamond electrode. Two irreversible oxidation peaks, a distinct one at +0.80 and a second poorly-defined one at +1.65 V, were observed when cyclic voltammetry was carried out in Britton-Robinson buffer solution at pH 7 (vs. Ag/AgCl). With optimized differential pulse voltammetric parameters (pulse amplitude 100 mV, pulse time 25 ms, step potential 5 mV and scan rate 10 mV s(-1)), the current response of yohimbine at +0.80 V was linearly proportional to the concentration in the range from 0.25 to 90.9 mu mol L-1 with a low detection limit of 0.13 mu mol L-1 (0.046 mg L-1) and a good repeatability (relative standard deviation of 2.5% at 18.4 mu mol L-1 for n = 6). The practical applicability of the developed method was demonstrated by the assessment of the total content of yohimbine in extracts of the primary bark of natural aphrodisiacs such as Pausinystalia yohimbe and Rauvolfia serpentina with recoveries in the range of 92-97%. The proposed electrochemical procedure represents an inexpensive and effective analytical alternative for the quality control analysis of products containing yohimbine and other biologically and structurally related alkaloids used as natural dietary supplements

    Boron-doped diamond film electrode as a sensitive and selective electrochemical sensor for the determination of paracetamol

    A simple, sensitive and selective differential pulse voltammetry method for determination of paracetamol on a bare (unmodified) boron-doped diamond film electrode has been developed. It was found by cyclic voltammetry that paracetamol provided the quasireversible wave with oxidation peak on the forward scan about +0.90 V and smaller reduction peak on the reverse scan at +0.68 V vs. Ag/AgCl. The effect of supporting electrolyte, pH and scan rate on voltammetric response of paracetamol was studied to select the optimum experimental conditions. The optimal conditions for quantification of paracetamol were obtained in acetate buffer solution at pH 5.0. The oxidation peak of paracetamol was chosen for evaluation and showed a systematic increase in peak current with increase of its concentration. Linear response of peak current on the concentration in the range from 2 × 10-7 to 6 × 10-5 mol L-1, good repeatability (RSD of 1.4% for ten successive measurements) and the detection limit of 1.1 × 10-7 mol L-1 were observed without any chemical modifications and electrochemical surface pretreatment of boron-doped diamond film electrode. The effect of possible interferents appeared to be negligible which evidently proved good selectivity. The practical analytical utility of proposed method was successfully demonstrated by determination of paracetamol in human urine samples and in pharmaceutical formulations (tablets) with results in a close statistical agreement to those declared by producer

    Abstract Wood fire residues can be found in various stages from less damaged material, pyrolysis products (a char) to deeply damaged material (ash). Moreover, wood burning can be influenced by fire accelerators and/or flame retardants. This paper provides a brief description and evaluation of conventional and comprehensive variants of analytical techniques with examples of their various applications at analyses of wood materials and wood fire residues. They include specificities of sampling, elemental chemical analysis, thermal analysis, extraction of compounds from fire debris, chromatographic methods and mass spectrometry, spectroscopic methods, chemometrics and quality assurance.</jats:p
