    Clinical validity assessment of a breast cancer risk model combining genetic and clinical information

    _Background:_ The extent to which common genetic variation can assist in breast cancer (BCa) risk assessment is unclear. We assessed the addition of risk information from a panel of BCa-associated single nucleotide polymorphisms (SNPs) on risk stratification offered by the Gail Model.

_Methods:_ We selected 7 validated SNPs from the literature and genotyped them among white women in a nested case-control study within the Women’s Health Initiative Clinical Trial. To model SNP risk, previously published odds ratios were combined multiplicatively. To produce a combined clinical/genetic risk, Gail Model risk estimates were multiplied by combined SNP odds ratios. We assessed classification performance using reclassification tables and receiver operating characteristic (ROC) curves. 

_Results:_ The SNP risk score was well calibrated and nearly independent of Gail risk, and the combined predictor was more predictive than either Gail risk or SNP risk alone. In ROC curve analysis, the combined score had an area under the curve (AUC) of 0.594 compared to 0.557 for Gail risk alone. For reclassification with 5-year risk thresholds at 1.5% and 2%, the net reclassification index (NRI) was 0.085 (Z = 4.3, P = 1.0×10^-5^). Focusing on women with Gail 5-year risk of 1.5-2% results in an NRI of 0.195 (Z = 3.8, P = 8.6×10^−5^).

_Conclusions:_ Combining clinical risk factors and validated common genetic risk factors results in improvement in classification of BCa risks in white, postmenopausal women. This may have implications for informing primary prevention and/or screening strategies. Future research should assess the clinical utility of such strategies.

    Variation in _PNPLA3_ is associated with outcomes in alcoholic liver disease

    Two recent genome-wide association studies have described associations of SNP variants in _PNPLA3_ with nonalcoholic fatty liver and plasma liver enzyme levels in population based cohorts. We investigated the contributions of these variants to clinical outcomes in Mestizo subjects with a history of excessive alcohol consumption. We show that non-synonymous variant rs738409[G] (I148M) in _PNPLA3_ is strongly associated with alcoholic liver disease and progression to alcoholic cirrhosis (unadjusted OR = 2.25, P = 1.7x10^-10^; ancestry-adjusted OR = 1.79, P = 1.9x10^-5^)

    The influence of coach turnover on student-athletes' affective states and team dynamics: An exploratory study in collegiate sports

    Coach turnover is frequent, especially in collegiate sports in the United States. To date, however, there is limited research on how coach turnover might affect student-athletes. Accordingly, we aimed to qualitatively explore how coach turnover might influence student-athletes’ psychosocial states and team dynamics in collegiate sports. Eleven athletic teams at a Midwestern Division I National Collegiate Athletic Association institution having undergone coach turnover during the past four years participated in the study. Four coaches and 21 student-athletes representing both individual and team sports, as well as two administrative staff members, participated in semi-structured interviews (coaches and administrators) and focus groups (student-athletes). Inductive data analysis revealed four first level themes: coach style and background, athlete affective states, team dynamics, and program culture. Our analysis suggested that, depending on their gender and coaching style, coaches might have a positive or negative impact on athletes’ affective states (e.g. positive and negative affect), team dynamics (e.g. cohesion; athlete leadership), and the program culture. Thus, practitioners might facilitate the positive impact of coaching turnover by assessing whether the new coach is a ‘good fit’ at the individual (micro), team (meso), and environmental (macro) level of analysis

    The forgotten kids: Experiences of potential student-athletes at a postsecondary football preparatory school

    The first preparatory institution was founded in 1635 to prepare elite men for public service, a role in the church, or admission to Harvard (Boyer, 1983). Nearly 400 years later, the objective of such institutions is no longer Harvard, but often an avenue for potential student-athletes (PSAs) to participate in collegiate sport (Thamel, 2007). The NCAA does not define nor regulate postgraduate preparatory institutions; however, Curran (2014) describes a preparatory institution (commonly referred to as prep schools) as a postgraduate institute that provides PSAs another year at a secondary institution prior to making the transition to college. Framed by Mincer’s (1958) model of Basic Human Capital, the purpose of this study is to examine the experiences of those attending or associated with one specific preparatory institution (X Academy). The first research question which explores prospective student-athletes choices to attend a preparatory institution revealed three final themes: (1) eligibility concerns, (2) athletic exposure and development, and (3) academic improvement.     Accounting for Ethics: Emphasis on Ethics Education in US Collegiate Business Curricula

    The accounting field holds a background in ethical understanding to be of great importance. The purpose of this project was to investigate the extent to which ethics education is emphasized and implemented within the business, and specifically accounting, curricula of US collegiate level institutions. Course curricula from a sample of 445 AACSB (Association to Advance Collegiate Schools of Business) accredited business programs in the US were examined for their emphasis on ethics education. This emphasis was quantified through a novel measurement, the Ethics Education Index, which is an originally constructed weighted measure of the number of stand-alone courses dealing with ethics in accounting and business programs as well as the frequency of ethical terminology used as descriptors within course catalog offerings and within each schools’ mission statement of the 2015-2016 course catalogs. From this data, the measures of the Ethics Education Index were standardized and then compared with various school categorical characteristics, including religious affiliation, number of undergraduate students and faculty, private vs. public institution, male or female dean, and US region, to be analyzed for any relationship between these variables. Prior hypotheses were made in relation to the correlations with the Ethics Education Index. Results and conclusions were drawn to determine the impact of the categorical variables on the Ethics Education Index score of the colleges and universities studied
