Scattered light mapping of protoplanetary disks
High-contrast scattered light observations have revealed the surface
morphology of several dozens of protoplanetary disks at optical and
near-infrared wavelengths. Inclined disks offer the opportunity to measure part
of the phase function of the dust grains that reside in the disk surface which
is essential for our understanding of protoplanetary dust properties and the
early stages of planet formation. We aim to construct a method which takes into
account how the flaring shape of the scattering surface of an (optically thick)
protoplanetary disk projects onto the image plane of the observer. This allows
us to map physical quantities (scattering radius and scattering angle) onto
scattered light images and retrieve stellar irradiation corrected (r^2-scaled)
images and dust phase functions. We apply the method on archival polarized
intensity images of the protoplanetary disk around HD 100546 that were obtained
with VLT/SPHERE in R'-band and VLT/NACO in H- and Ks-band. The brightest side
of the r^2-scaled R'-band polarized intensity image of HD 100546 changes from
the far to the near side of the disk when a flaring instead of a geometrically
flat disk surface is used for the r^2-scaling. The decrease in polarized
surface brightness in the scattering angle range of ~40-70 deg is likely a
result of the dust phase function and degree of polarization which peak in
different scattering angle regimes. The derived phase functions show part of a
forward scattering peak which indicates that large, aggregate dust grains
dominate the scattering opacity in the disk surface. Projection effects of a
protoplanetary disk surface need to be taken into account to correctly
interpret scattered light images. Applying the correct scaling for the
correction of stellar irradiation is crucial for the interpretation of the
images and the derivation of the dust properties in the disk surface layer.Comment: Accepted for publication in A&A, 6 pages, 3 figure

Generic sample preparation combined with high-resolution liquid chromatography- time-of-flight mass spectrometry for unification of urine screening in doping-control laboratories
A unification of doping-control screening procedures of prohibited small molecule substances—including stimulants, narcotics, steroids, ß2-agonists and diuretics—is highly urgent in order to free resources for new classes such as banned proteins. Conceptually this may be achieved by the use of a combination of one gas chromatography–time-of-flight mass spectrometry method and one liquid chromatography–time-of-flight mass spectrometry method. In this work a quantitative screening method using high-resolution liquid chromatography in combination with accurate-mass time-of-flight mass spectrometry was developed and validated for determination of glucocorticosteroids, ß2-agonists, thiazide diuretics, and narcotics and stimulants in urine. To enable the simultaneous isolation of all the compounds of interest and the necessary purification of the resulting extracts, a generic extraction and hydrolysis procedure was combined with a solid-phase extraction modified for these groups of compounds. All 56 compounds are determined using positive electrospray ionisation with the exception of the thiazide diuretics for which the best sensitivity was obtained by using negative electrospray ionisation. The results show that, with the exception of clenhexyl, procaterol, and reproterol, all compounds can be detected below the respective minimum required performance level and the results for linearity, repeatability, within-lab reproducibility, and accuracy show that the method can be used for quantitative screening. If qualitative screening is sufficient the instrumental analysis may be limited to positive ionisation, because all analytes including the thiazides can be detected at the respective minimum required levels in the positive mode. The results show that the application of accurate-mass time-of-flight mass spectrometry in combination with generic extraction and purification procedures is suitable for unification and expansion of the window of screening methods of doping laboratories. Moreover, the full-scan accurate-mass data sets obtained still allow retrospective examination for emerging doping agents, without re-analyzing the samples
Inheritance of determinants of flower colour in tetraploid roses
The choice of selection breeding for crop improvement in cut roses requires a better understanding of biological mechanisms and knowledge of the inheritance of the major target traits which can lead to new or improved screening methods. Colour is still the most important trait in cut roses. A tetraploid mapping population will be characterized for flower colour, by using colour charts such as the official chart of the Royal Horticultural Society, and additionally, by image analysis and measuring reflectance using a spectrocolorimeter. The genetics of flower colour will be studied. In addition, flower petals of all genotypes will be analysed by HPLC to characterize secondary metabolic components that determine flower colour, such as anthocyanins. The inheritance of these components will also be assessed and compared to that of flower colour. Preliminary results show that the most effective method to quantify colour is by HPLC analysis of the extracted anthocyanins. The highest pelargonidin concentrations occur at relatively low cyanidin concentrations. PynPoint: a modular pipeline architecture for processing and analysis of high-contrast imaging data
The direct detection and characterization of planetary and substellar
companions at small angular separations is a rapidly advancing field. Dedicated
high-contrast imaging instruments deliver unprecedented sensitivity, enabling
detailed insights into the atmospheres of young low-mass companions. In
addition, improvements in data reduction and PSF subtraction algorithms are
equally relevant for maximizing the scientific yield, both from new and
archival data sets. We aim at developing a generic and modular data reduction
pipeline for processing and analysis of high-contrast imaging data obtained
with pupil-stabilized observations. The package should be scalable and robust
for future implementations and in particular well suitable for the 3-5 micron
wavelength range where typically (ten) thousands of frames have to be processed
and an accurate subtraction of the thermal background emission is critical.
PynPoint is written in Python 2.7 and applies various image processing
techniques, as well as statistical tools for analyzing the data, building on
open-source Python packages. The current version of PynPoint has evolved from
an earlier version that was developed as a PSF subtraction tool based on PCA.
The architecture of PynPoint has been redesigned with the core functionalities
decoupled from the pipeline modules. Modules have been implemented for
dedicated processing and analysis steps, including background subtraction,
frame registration, PSF subtraction, photometric and astrometric measurements,
and estimation of detection limits. The pipeline package enables end-to-end
data reduction of pupil-stabilized data and supports classical dithering and
coronagraphic data sets. As an example, we processed archival VLT/NACO L' and
M' data of beta Pic b and reassessed the planet's brightness and position with
