The Role of Muscular TDP-43 in Neuromuscular Junction Development and Progress of ALS

Amyotrophic lateral sclerosis (ALS) is a fatal disease with fast progression. Patients usually die three to five years after diagnosis. The main characteristics are progressive neurodegeneration, following muscular weakness, and atrophy. The most common cause of death is respiratory muscle failure. Studies have shown that the highlight of the disease is TDP-43 inclusions in neuronal cytoplasm. The symptoms of ALS can be recapitulated in Drosophila melanogaster. The Drosophila‘s ortholog is called TBPH. TBPH knockout flies show locomotion problems, shorter lifespan, and anatomical changes in the neuromuscular junction (NMJ). Our studies aimed to reveal the function of TBPH in muscles. We took advantage of RNA interference and silenced the protein exclusively in muscles. Moreover, we expressed TBPH in tbphΔ23/- fly and tried to understand whether it can rescue reduced mobility, lifespan, and anatomical properties of NMJ. We observed changes in lifespan, motility, and NMJ structure when TBPH was silenced in muscles. Expression of the protein in tbphΔ23/- Drosophila completely recovered the motility of larvae and the level of proteins located in the postsynaptic compartment of NMJ. One of the proteins was Disc large (Dlg), known as scaffold protein involved in NMJ development and maintenance of NMJ structure. Expression of Dlg recovered the phenotype in terms of mobility, lifespan, and NMJ formation. We also demonstrated that the drop of Dlg levels could be seen in neuroblastoma cells and differentiated iPS cells of ALS patients. With that been discovered, not only have we confirmed the relevance of our studies, but we have also opened a new possibility for drug development and treatment. Moreover, with the research, we contributed to a recent hypothesis that muscles can be involved in the onset of ALS

National universities creation (2007-2015): reshaping of the Argentinian higher education system

La pregunta ¿más o menos universidades en nuestro país? ha circulado en los últimos años en diferentes espacios de discusión. El presente artículo da cuenta del impacto social que ha tenido el proceso de creación de dieciséis nuevas universidades nacionales de gestión estatal de 2007 a 2015, y que da como resultado la configuración de un nuevo sistema nacional de educación superior en la Argentina. Se busca desde el artículo refutar algunas interpretaciones y falsas comparaciones que intentan explicar el sistema universitario nacional como excesivo, desmedido e irracional. Para contrastar estos argumentos se exploran algunas experiencias que finalmente reflejan un hecho inclusivo sin precedentes que abre el camino a nuevas realidades, nuevos derechos y nuevas posibilidades para un importante sector de la población, especialmente aquellos que habitan en el conurbano bonaerense.The question, whether more or less universities in our country has circulated during the last years in different discussion spaces. The current article gives an account of the social impact caused by the process of creation of sixteen new national state universities during the years 2007 and 2015, which has as a result the configuration of a new social system of higher education in Argentina. From this article, the intention is to refute some interpretations and false comparisons that attempt to consider the national university system excessive, disproportionate and irrational. In order to counter these arguments, some experiences are explored, which show an inclusive unprecedented event that opens the way to new realities, new rights and new possibilities for an important sector of the population, especially those who inhabit the suburbs around Buenos Aires city.Facultad de Periodismo y Comunicación Socia
