Connecting instructional and cognitive aspects of an LE: A study of the global seminar project
My research problem is based on the lack of unifying conceptual cohesion between the discourses concerning cognitive and instructional aspects of learning environments (LE). I contrast that lack with practical developments of LE studies connected at the level of practical implementation and evaluation. Next, I briefly review the LE boundaries, which are established within cognitive and instructional domains of LE research. This study aims to bring the aforementioned discourses together via reconsidering and clarifying LE conceptual understandings. Using the case of a global seminar project (GSP), my goal is to define an LE as a conceptual phenomenon and a practical model with the two objectives of determining the GSP's components that establish the course in a context of higher education; and the GSP's specific contributions to deep learning. Within a qualitative research framework, I used three forms of data collection: 20 open-ended interviews of the GSP instructors; 11 direct observations of the GSP classroom; and the GSP's written documents and artifacts. Noting the contextual significance of the suggested LE framework that resulted from this study, I propose a definition of an LE as a conceptual phenomenon and educational model. Here, I introduce the key understandings of an LE and outline its significance based on a broader analysis of the results. Remote Methods of Conducting Transactions Using Digital Technologies
Objective: to substantiate the need to identify new contractual constructs (models) taking into account the specific relations associated with the use of remote method of contract conclusion through digital technologies and to study the possible risks for their participants.Methods: along with special legal methods, the method of critical analysis was fundamental for the research process, which allowed us to evaluate and interpret the main sources and norms of civil law in relation to distant transactions. It also allowed assessing the current state of legislation in this area in the context of developing processes of digitalization and technologization of civil-law relations.Results: a critical analysis of the current state of legal regulation of remote ways of concluding contracts is presented, their classification is given. It is concluded that the digital technologies development gives rise to new remote ways of transactions, as well as fills with new content the procedures of contract conclusion, traditional for civil law. The expediency of singling out the concept of a “distant transaction” as a legal category in order to create a special civil-law regime is substantiated, and the basic concept being that of a “distant contract”. Certain types of distant contracts are analyzed to substantiate the need for special legal regimes in cases when the distant method of contract conclusion is combined with the use of digital technologies. It poses such problems as the distribution of risks of technological failures, hacker attacks, compliance with the balance of interests of the parties taking into account information asymmetry, and the need to protect the weaker party.Scientific novelty: an attempt is made to define such concepts as a “distant contract” and a “distant transaction” and to identify their features. The expediency is substantiated of considering a distant contract as a separate legal construction (model) of the contract. Within this framework, a special legal regime should be developed and fixed, which can be extended to unilateral distant transactions. The problems of legal regulation caused by the use of information technologies are formulated, and legal constructions for their solution are proposed.Practical significance: the final conclusions and proposals can be used both in contractual practice by the participants of civil turnover and for the normative consolidation of the concept and features of “distant contract”, “distant transaction”. A special legal regime can be created, taking into account the specificity generated by the use of digital technologies
Gene expression analysis of TIL rich HPV-driven head and neck tumors reveals a distinct B-cell signature when compared to HPV independent tumors
Human papilloma virus (HPV)-associated head and neck squamous cell carcinoma (HNSCC) has a better prognosis than it's HPV negative (HPV(-)) counterpart. This may be due to the higher numbers of tumor-infiltrating lymphocytes (TILs) in HPV positive (HPV(+)) tumors. RNA-Sequencing (RNA-Seq) was used to evaluate whether the differences in clinical behaviour simply reflect a numerical difference in TILs or whether there is a fundamental behavioural difference between TILs in these two settings. Thirty-nine HNSCC tumors were scored for TIL density by immunohistochemistry. After the removal of 16 TILlow tumors, RNA-Seq analysis was performed on 23 TILhigh/med tumors (HPV(+) n=10 and HPV(-) n=13). Using EdgeR, differentially expressed genes (DEG) were identified. Immune subset analysis was performed using Functional Analysis of Individual RNA-Seq/ Microarray Expression (FAIME) and immune gene RNA transcript count analysis. In total, 1,634 DEGs were identified, with a dominant immune signature observed in HPV(+) tumors. After normalizing the expression profiles to account for differences in B- and T-cell number, 437 significantly DEGs remained. A B-cell associated signature distinguished HPV(+) from HPV(-) tumors, and included the DEGs CD200, GGA2, ADAM28, STAG3, SPIB, VCAM1, BCL2 and ICOSLG; the immune signal relative to T-cells was qualitatively similar between TILs of both tumor cohorts. Our findings were validated and confirmed in two independent cohorts using TCGA data and tumor-infiltrating B-cells from additional HPV(+) HNSCC patients. A B-cell associated signal segregated tumors relative to HPV status. Our data suggests that the role of B-cells in the adaptive immune response to HPV(+) HNSCC requires re-assessment
Plant virus particles carrying tumour antigen activate TLR7 and induce high levels of protective antibody
Induction of potent antibody is the goal of many vaccines targeted against infections or cancer. Modern vaccine designs that use virus-like particles (VLP) have shown efficacy for prophylactic vaccination against virus-associated cancer in the clinic. Here we used plant viral particles (PVP), which are structurally analogous to VLP, coupled to a weak idiotypic (Id) tumour antigen, as a conjugate vaccine to induce antibody against a murine B-cell malignancy. The Id-PVP vaccine incorporates a natural adjuvant, the viral ssRNA, which acts via TLR7. It induced potent protective anti-Id antibody responses in an in vivo mouse model, superior to the "gold standard" Id vaccine, with prevalence of the IgG2a isotype. Combination with alum further increased antibody levels and maintained the IgG2a bias. Engagement of TLR7 in vivo was followed by secretion of IFN-? by plasmacytoid dendritic cells and by activation of splenic CD11chi conventional dendritic cells. The latter was apparent from up-regulation of co-stimulatory molecules and from secretion of a wide range of inflammatory cytokines and chemokines including the Th1-governing cytokine IL-12, in keeping with the IgG2a antibody isotype distribution. PVP conjugates are a novel cancer vaccine design, offering an attractive molecular form, similar to VLP, and providing T-cell help. In contrast to VLP, they also incorporate a safe "in-built" ssRNA adjuvant
Radial-piston pump for drive of test machines
The article reviews the development of radial-piston pump with phase control and alternating-flow mode for seismic-testing platforms and other test machines. The prospects for use of the developed device are proved. It is noted that the method of frequency modulation with the detection of the natural frequencies is easily realized by using the radial-piston pump. The prospects of further research are given proof

Specifics of Digital Interaction of Regional Mass Media with Audience: Nizhny Novgorod Experience
The focus of the authors of the article attention is the analysis of the practical experience of mastering digital media technologies aimed at the formation and strengthening of constructive interaction with the readership. The subject of analysis was the sites and social networks of traditional mass media of the closed administrative-territorial formation (ZATO) of Sarov (Nizhegorod region). These sources were studied in terms of the specifics of digital content distribution, the formation of new technological approaches to interaction with the audience, and the search for creative ways of establishing constructive interaction with the reader. It has been established that the use of digital technologies in the local information market has not yet become a universal trend. At the same time, it is noted that the analysis of the activity of regional mass media in social networks, video hosting, messengers revealed positive practices of interaction with audience groups, in particular, a clearly noticeable tendency to establish interactive communication with the target audience. However, conclusions are drawn about the insufficient use of available communication resources in building a mutually interested dialogue, which in the future may negatively affect the prospective development of the local press, lead to stagnation due to inattention to the requests and expectations of the audience
Prognostic impact of clinical and genetic factors on delayed elimination of methotrexate in children with acute lymphoblastic leukemia
Background. Methotrexate (MTX) in high doses (1000–5000 mg/m2), occupies one of the leading places in modern programs of therapy of acute lymphoblastic leukemia (ALL) in children. Achievement of high long-term survival rates in children with ALL has become possible thanks to this drug. However, no less important problem is the toxicity of the treatment and prediction of its efficacy and safety, in this regard, the role of pharmacogenetic studies in the identification of polymorphisms in candidate genes affecting the pharmacokinetics of MTX is increasing.Objective. To determine predictors of delayed elimination of methotrexate using pharmacokinetic biomarkers in children with acute lymphoblastic leukemia.Materials and methods. We prospectively analyzed the database of pediatric patients with ALL within the framework of an observational (cohort) singlecenter study. The study included 124 children diagnosed with ALL who received therapy according to the ALL-IC BFM 2009 protocol including high-dosed MTX. Real-time PCR method was used to study polymorphisms of ABCB1 and SLCO1B1 genes. The study material was peripheral blood. Statistical analysis ofpharmacogenetic biomarkers influence on toxicity and efficacy of therapy was performed using SPSS Statistics 26.0 program (USA). To form mathematical prognostic models, we used the method of logistic function construction using binary logistic regression with step-by-step selection of factors and, if necessary, additional construction of ROC-curves with subsequent ROC-analysis. Differences were considered significant at p < 0.05; at p ≥ 0.05, differences were considered unlikely and statistically insignificant.Results. Based on the results of the conducted complex analysis of efficacy and safety of high-dosed MTX therapy, a reliable (p < 0.001) prognostic model with high sensitivity, specificity and efficacy (>70 %, respectively) was developed, demonstrating the interrelationships of clinical and genetic factors influencing the delay of MTX elimination in children with ALL, which confirms the necessity of implementing pharmacogenetic testing in real clinical practice.Conclusion. Determination of polymorphisms of genes providing transport and metabolism of cytostatics should be used in practical work of oncohematological clinics for individualization of therapy and ensuring its safety
