    mRNA localization in the Drosophila germline.

    Localization and the associated translational control of mRNA is a well established mechanism for segregating cellular protein expression. Drosophila has been instrumental in deciphering the prevailing mechanisms of mRNA localization and regulation. This review will discuss the diverse roles of mRNA localization in the Drosophila germline, the cis-elements and cellular components regulating localization and the superimposition of translational regulatory mechanisms. Despite a history of discovery, there are still many fundamental questions regarding mRNA localization that remain unanswered. Take home messages, outstanding questions and future approaches that will likely lead to resolving these unknowns in the future are summarized at the end.This work was supported by the University of Cambridge, ISSF to T.T.W.     From Bare Metal to Virtual: Lessons Learned when a Supercomputing Institute Deploys its First Cloud

    As primary provider for research computing services at the University of Minnesota, the Minnesota Supercomputing Institute (MSI) has long been responsible for serving the needs of a user-base numbering in the thousands. In recent years, MSI---like many other HPC centers---has observed a growing need for self-service, on-demand, data-intensive research, as well as the emergence of many new controlled-access datasets for research purposes. In light of this, MSI constructed a new on-premise cloud service, named Stratus, which is architected from the ground up to easily satisfy data-use agreements and fill four gaps left by traditional HPC. The resulting OpenStack cloud, constructed from HPC-specific compute nodes and backed by Ceph storage, is designed to fully comply with controls set forth by the NIH Genomic Data Sharing Policy. Herein, we present twelve lessons learned during the ambitious sprint to take Stratus from inception and into production in less than 18 months. Important, and often overlooked, components of this timeline included the development of new leadership roles, staff and user training, and user support documentation.     Site-selective protein modification via disulfide rebridging for fast tetrazine/trans-cyclooctene bioconjugation

    An inverse electron demand Diels–Alder reaction between tetrazine and trans-cyclooctene (TCO) holds great promise for protein modification and manipulation. Herein, we report the design and synthesis of a tetrazine-based disulfide rebridging reagent, which allows the site-selective installation of a tetrazine group into disulfide-containing peptides and proteins such as the hormone somatostatin (SST) and the antigen binding fragment (Fab) of human immunoglobulin G (IgG). The fast and efficient conjugation of the tetrazine modified proteins with three different TCO-containing substrates to form a set of bioconjugates in a site-selective manner was successfully demonstrated for the first time. Homogeneous, well-defined bioconjugates were obtained underlining the great potential of our method for fast bioconjugation in emerging protein therapeutics. The formed bioconjugates were stable against glutathione and in serum, and they maintained their secondary structure. With this work, we broaden the scope of tetrazine chemistry for site-selective protein modification to prepare well-defined SST and Fab conjugates with preserved structures and good stability under biologically relevant conditions

    R\^ole of the pion electromagnetic form factor in the Δ(1232)γN\Delta(1232) \to \gamma^\ast N timelike transition

    The Δ(1232)γN\Delta(1232) \to \gamma^\ast N magnetic dipole form factor (GMG_M^\ast) is described here within a new covariant model that combines the valence quark core together with the pion cloud contributions. The pion cloud term is parameterized by two terms: one connected to the pion electromagnetic form factor, the other to the photon interaction with intermediate baryon states. The model can be used in studies of pp and heavy ion collisions. In the timelike region this new model improves the results obtained with a constant form factor model fixed at its value at zero momentum transfer. At the same time, and in contrast to the Iachello model, this new model predicts a peak for the transition form factor at the expected position, i.e. at the ρ\rho mass pole. We calculate the decay of the ΔγN\Delta \to \gamma N transition, the Dalitz decay (Δe+eN\Delta \to e^+ e^- N), and the Δ\Delta mass distribution function. The impact of the model on dilepton spectra in pp collisions is also discussed.Comment: 11 pages, 10 figure

    Electrically detected magnetic resonance of carbon dangling bonds at the Si-face 4H-SiC/SiO2_2 interface

    SiC based metal-oxide-semiconductor field-effect transistors (MOSFETs) have gained a significant importance in power electronics applications. However, electrically active defects at the SiC/SiO2_2 interface degrade the ideal behavior of the devices. The relevant microscopic defects can be identified by electron paramagnetic resonance (EPR) or electrically detected magnetic resonance (EDMR). This helps to decide which changes to the fabrication process will likely lead to further increases of device performance and reliability. EDMR measurements have shown very similar dominant hyperfine (HF) spectra in differently processed MOSFETs although some discrepancies were observed in the measured gg-factors. Here, the HF spectra measured of different SiC MOSFETs are compared and it is argued that the same dominant defect is present in all devices. A comparison of the data with simulated spectra of the C dangling bond (PbC_\textrm{bC}) center and the silicon vacancy (VSi_\textrm{Si}) demonstrates that the PbC_\textrm{bC} center is a more suitable candidate to explain the observed HF spectra.Comment: Accepted for publication in the Journal of Applied Physic
