    Mechanism of nitrogen metabolism-related parameters and enzyme activities in the pathophysiology of autism

    <p>Abstract</p> <p>Background</p> <p>There is evidence that impaired metabolism play an important role in the etiology of many neuropsychiatric disorders. Although this has not been investigated to date, several recent studies proposed that nitrogen metabolism-related parameters may have a pathophysiological role in autism.</p> <p>Methods</p> <p>The study enrolled 20 Saudi boys with autism aged 4 to 12 years and 20 healthy controls matched for age and gender. Levels of creatine, urea, ammonia, gamma-aminobutyric acid (GABA), glutamate:glutamine (Glu:Gln) ratio, and enzymatic activities of glutamate dehydrogenase, 5'-nucleotidase, and adenosine deaminase (ADA) were determined in plasma samples from both groups.</p> <p>Results</p> <p>We found a significant elevation of creatine, 5'-nucleotidase, GABA, and glutamic acid and a significant decrease in the enzymatic activity of ADA and glutamine level in patients with autism compared with healthy controls. The most significant variation between the two groups was found in the Glu:Gln ratio.</p> <p>Conclusion</p> <p>A raised Glu:Gln ratio together with positive correlations in creatine, GABA, and 5'-nucleotidase levels could contribute to the pathophysiology of autism, and might be useful diagnostic markers. The mechanism through which these parameters might be related to autism is discussed in detail.</p

    Analysis and design of elevated liquid-filled reinforced concrete conical tanks

    Reinforced concrete conical tanks are used in municipalities and industrial applications as liquid containing vessels. These tanks can be ground supported tanks or elevated on a supporting shaft. Although most design codes provide guidelines for rectangular and cylindrical tanks, no guidance is provided in these codes for conical tanks. This study focuses on assessing the accuracy of a design approach based on the provisions of Portland Cement Association Circular Concrete Tanks without Pre-stressing code for cylindrical tanks combined with an equivalent cylinder approach provided by the American Water Works Association-D100. Several reinforced concrete conical tanks with different geometric configurations are analyzed under the effect of hydrostatic loading using this approach. The internal forces obtained according to code provisions are compared with those predicted by a linear finite element analysis model developed for all studied tanks. The finite element analysis model is based on a degenerated consistent sub-parametric triangular shell element developed in-house. The results of this comparison show that Portland Cement Association Circular Concrete Tanks without Pre-stressing approach leads to larger hoop tension and smaller meridional moment compared to those obtained from the finite element analysis. Therefore, the Portland Cement Association Circular Concrete Tanks without Pre-stressing provisions combined with the equivalent cylinder approach lead to an inadequate design if applied to conical-shaped tanks. </jats:p

    The Berkeley Puppet Interview: A screening instrument for measuring psychopathology in young children

    Item does not contain fulltextWhile child self-reports of psychopathology are increasingly accepted, little standardized instruments are utilized for these practices. The Berkeley Puppet Interview (BPI) is an age-appropriate instrument for self-reports of problem behavior by young children. Psychometric properties of the Dutch version of the BPI will be reported, specifically, test-retest reliability, intra-class correlations, congruent and concurrent validity. In a sample of 300 children (M (age) = 7.04 years, SD = 1.15), the BPI was administered twice, with a 1-year interval. Parents and teachers filled out questionnaires about their children's problem behavior. Findings from the analyses indicate that the BPI subscales have sufficient test-retest reliability and can be reliably coded. Furthermore, findings suggest adequate congruent validity. More support for concurrent validity is found among externalizing problems in comparison to internalizing problems. With regard to the present study, the BPI seems to have adequate psychometric properties. As such, the BPI enables interviewing young children about their psychopathology-related symptoms in a standardized way. The BPI could be applied in clinical practice as a complement to the diagnostic cycle, allowing children's self-reports to play an increasingly important role.14 p

    Molluscicidal activity of Hammada scoparia (Pomel) Iljin leaf extracts and the principal alkaloids isolated from them against Galba truncatula

    The molluscicidal activity of Hammada scoparia leaf extracts and the principal alkaloids isolated from them (carnegine and N-methylisosalsoline) were tested against the mollusc gastropod, Galba truncatula, the intermediate host of Fasciola hepatica in Tunisia. The results indicated that the molluscicidal activity was correlated with the presence of alkaloids. A significant molluscicidal value, according to the World Health Organization, was found with the methanol extract (LC50 = 28.93 ppm). Further fractionation of the methanolic extract led to the isolation of two principal alkaloids: carnegine and N-methylisosalsoline. These alkaloids are isoquinolines that have not previously been characterised for their molluscicidal activity. The N-methylisosalsoline possesses the highest molluscicidal activity (LC50 = 0.47 &#956;M against G. truncatula)

    Pharmacological Modulation of GABA Function in Autism Spectrum Disorders: A Systematic Review of Human Studies

    Autism spectrum disorders are an emerging health problem worldwide, but little is known about their pathogenesis. It has been hypothesized that autism may result from an imbalance between excitatory glutamatergic and inhibitory GABAergic pathways. Commonly used medications such as valproate, acamprosate, and arbaclofen may act on the GABAergic system and be a potential treatment for people with ASD. The present systematic review aimed at evaluating the state-of-the-art of clinical trials of GABA modulators in autism. To date there is insufficient evidence to suggest the use of these drugs in autistic subjects, even if data are promising. Of note, short-term use of all the reviewed medications appears to be safe. Future well designed trials are needed to elucidate these preliminary findings
