    Evolutionary relationships among barley and <i>Arabidopsis</i> core circadian clock and clock-associated genes

    The circadian clock regulates a multitude of plant developmental and metabolic processes. In crop species, it contributes significantly to plant performance and productivity and to the adaptation and geographical range over which crops can be grown. To understand the clock in barley and how it relates to the components in the Arabidopsis thaliana clock, we have performed a systematic analysis of core circadian clock and clock-associated genes in barley, Arabidopsis and another eight species including tomato, potato, a range of monocotyledonous species and the moss, Physcomitrella patens. We have identified orthologues and paralogues of Arabidopsis genes which are conserved in all species, monocot/dicot differences, species-specific differences and variation in gene copy number (e.g. gene duplications among the various species). We propose that the common ancestor of barley and Arabidopsis had two-thirds of the key clock components identified in Arabidopsis prior to the separation of the monocot/dicot groups. After this separation, multiple independent gene duplication events took place in both monocot and dicot ancestors.     ERBB2 in Cat Mammary Neoplasias Disclosed a Positive Correlation between RNA and Protein Low Expression Levels: A Model for erbB-2 Negative Human Breast Cancer

    Human ERBB2 is a proto-oncogene that codes for the erbB-2 epithelial growth factor receptor. In human breast cancer (HBC), erbB-2 protein overexpression has been repeatedly correlated with poor prognosis. In more recent works, underexpression of this gene has been described in HBC. Moreover, it is also recognised that oncogenes that are commonly amplified or deleted encompass point mutations, and some of these are associated with HBC. In cat mammary lesions (CMLs), the overexpression of ERBB2 (27%–59.6%) has also been described, mostly at the protein level and although cat mammary neoplasias are considered to be a natural model of HBC, molecular information is still scarce. In the present work, a cat ERBB2 fragment, comprising exons 10 to 15 (ERBB2_10–15) was achieved for the first time. Allelic variants and genomic haplotype analyses were also performed, and differences between normal and CML populations were observed. Three amino acid changes, corresponding to 3 non-synonymous genomic sequence variants that were only detected in CMLs, were proposed to damage the 3D structure of the protein. We analysed the cat ERBB2 gene at the DNA (copy number determination), mRNA (expression levels assessment) and protein levels (in extra- and intra protein domains) in CML samples and correlated the last two evaluations with clinicopathological features. We found a positive correlation between the expression levels of the ERBB2 RNA and erbB-2 protein, corresponding to the intracellular region. Additionally, we detected a positive correlation between higher mRNA expression and better clinical outcome. Our results suggest that the ERBB2 gene is post-transcriptionally regulated and that proteins with truncations and single point mutations are present in cat mammary neoplastic lesions.     Bioactive compounds as potential angiotensin-converting enzyme II inhibitors against COVID-19: a scoping review

    Objective and design The current study aimed to summarize the evidence of compounds contained in plant species with the ability to block the angiotensin-converting enzyme 2 (ACE-II), through a scoping review. Methods PubMed and Scopus electronic databases were used for the systematic search and a manual search was performed Results Studies included were characterized as in silico. Among the 200 studies retrieved, 139 studies listed after the exclusion of duplicates and 74 were included for the full read. Among them, 32 studies were considered eligible for the qualitative synthesis. The most evaluated class of secondary metabolites was flavonoids with quercetin and curcumin as most actives substances and terpenes (isothymol, limonin, curcumenol, anabsinthin, and artemisinin). Other classes that were also evaluated were alkaloid, saponin, quinone, substances found in essential oils, and primary metabolites as the aminoacid l-tyrosine and the lipidic compound 2-monolinolenin. Conclusion This review suggests the most active substance from each class of metabolites, which presented the strongest affinity to the ACE-II receptor, what contributes as a basis for choosing compounds and directing the further experimental and clinical investigation on the applications these compounds in biotechnological and health processes as in COVID-19 pandemic

    Desigualdades na autoavaliacao da saude bucal em adultos

    OBJETIVO Analisar a associação entre autoavaliação da saúde bucal em adultos e desigualdades sociodemográficas. MÉTODOS Estudo transversal com 2.016 adultos de 20 a 59 anos de idade, de Florianópolis, SC, em 2009. A amostra foi obtida por duplo estágio (setores censitários e domicílios). Os dados foram coletados por entrevistas domiciliares face a face. O desfecho foi autoavaliação da saúde bucal. As variáveis exploratórias foram caracterizadas em blocos demográficos, socioeconômicos, de utilização de serviços e de condições bucais autorreferidas. Foi realizada análise de regressão multivariável de Poisson e estimadas as razões de prevalências e respectivos intervalos de 95% de confiança. RESULTADOS A prevalência de autoavaliação negativa da saúde bucal foi de 33,2% (IC95% 29,8;36,6). Idade avançada, referir-se como pardo, possuir menor escolaridade, ter consultado o dentista há três anos ou mais, ter realizado a última consulta em consultório público, possuir menos de dez dentes naturais presentes em pelo menos um arco, perceber necessidade de tratamento odontológico, apresentar sensação de boca seca e dificuldade de alimentação em virtude dos dentes foram associados à autoavaliação negativa da saúde bucal na análise ajustada. CONCLUSÕES A autoavaliação da saúde bucal reflete as desigualdades em saúde e está relacionada às piores condições socioeconômicas, menor uso de serviços de saúde e pior condição bucal autorreferida

    Percepções da família da criança com doença crônica frente às relações com profissionais da saúde

    A prevalência de doença crônica na infância tem aumentado, impondo à família contato contínuo com profissionais de saúde, ocasião em que a relação desempenha papel relevante. Raras explorações científicas abordam tal temática, apesar de ser apontada como núcleo fundamental para a interação efetiva. O objetivo desse estudo foi caracterizar como a família de criança doente crônica percebe sua relação com os profissionais de saúde. Foram entrevistados 20 cuidadores de criança com doença crônica. O referencial teórico e metodológico adotados foram, respectivamente, o Interacionismo Simbólico e a análise de conteúdo temática. O processo analítico revelou quatro categorias temáticas: papel do cuidador, relações angustiantes, serviços e profissionais e buscando relações acolhedoras. O resultado deste processo interacional que apresenta incongruência de expectativas e comportamentos entre profissional e família é falho e gera sentimento de desamparo, o qual é amenizado pela escuta e compreensão de outras famílias em situação similar

    Biflorin: an o-naphthoquinone of clinical significance

    Biflorin is an o-naphthoquinone with proven cytotoxic effects on tumor cells showing antimicrobial, antitumor and antimutagenic activities. Biflorin is an isolated compound taken from the roots of the plant Capraria biflora L. (Schrophulariaceae), indigenous of the West Indies and South America, which is located in temperate or tropical areas. This compound has shown to be strongly active against grampositive and alcohol-acid-resistant bacteria. It has been efficient in inhibiting the proliferation tumor cell lines CEM, HL-60, B16, HCT-8 and MCF-7. Recently, SK-Br3 cell line was treated with biflorin showing important cytotoxic effects. In this article, information related to the first structural characterization studies are presented, as well as the latest reports concerning the biological activity of this molecule
