    Inconsistencies in Reported Employment Characteristics among Employed Stayers

    The paper deals with measurement error, and its potentially distorting role, in information on industry and professional status collected by labour force surveys. The focus of our analyses is on inconsistent information on these employment characteristics resulting from yearly transition matrices for workers who were continuously employed over the year and who did not change job. As a case-study we use yearly panel data for the period from April 1993 to April 2003 collected by the Italian Quarterly Labour Force Survey. The analysis goes through four steps: (i) descriptive indicators of (dis)agreement; (ii) testing whether the consistency of repeated information significantly increases when the number of categories is collapsed; (iii) examination of the pattern of inconsistencies among response categories by means of Goodman's quasi-independence model; (iv) comparisons of alternative classifications. Results document sizable measurement error, which is only moderately reduced by more aggregated classifications.     The transition from pediatric to adult age in patients with Dravet Syndrome: A study on the current management pathways and the challenges ahead

    Dravet Syndrome (DS) is an infantile onset drug-resistant developmental and epileptic encephalopathy, often associated with comorbidities including intellectual disability, behavioral disorders and gait problems. Epileptic seizures usually begin in the first year of life and persist into adulthood, resistant to antiseizure medications. Within the complex care pathway faced by these patients and their families, the transition from pediatric care centers to adult centers represents a critical phase of the patient's journey. If not managed optimally, transition can have significant repercussions on the quality of care and patient management. We conducted a mixedmethods investigation, involving a quantitative survey of 54 neurologists and neuropsychiatrists from 28 national reference centers, and a qualitative survey with 5 specialists and 3 caregivers of patients with DS. The study aimed to investigate the transition of patients from child neurologist clinics to adult clinics exploring management methods, challenges, and unmet needs of patients and their caregivers. The results indicate that transitions to adult centers are currently heterogeneous and often unstructured, with significant implications in terms of the quality of care provided. Particularly, structured transition programs are available in only 70% of centers, and many patients remain in pediatric care beyond adulthood due to perceived better care quality and stability concerns. Additionally, around 19% of patients abandon adult care centers within a year of transition. In order to improve the quality of care and the quality of life for patients with this syndrome, it is essential to listen to the requests of their caregivers, including continuity in patient management, psychological support and planning for the future of their loved ones. The overall fragmented nature of transition strategies highlights the need for initiatives to provide greater structure and continuity in care pathways

    Stable ubiquitin conjugation for biological interrogation of ubiquitinated tau repeat domain

    Protein semisynthesis approaches are key for gaining insights into the effects of post-translational modifications (PTMs) on the structure and function of modified proteins. Among PTMs, ubiquitination involves the conjugation of a small protein modifier to a substrate amino acid residue and is unique in controlling a variety of cellular processes. Interest has grown in understanding the role of ubiquitination in neurodegenerative conditions, including tauopathies. The latter are characterized by the accumulation of the intrinsically disordered protein tau in the form of neurofibrillary tangles in the brains of patients. The presence of ubiquitinated tau in the pathological aggregates suggests that ubiquitination might play a role in the formation of abnormal protein deposits. In this study, we developed a new strategy, based on dehydroalanine chemistry, to install wild type ubiquitin on a tau repeat domain construct with site-specificity. We optimized a three-step reaction which yielded a good amount of highly pure tau repeat domain ubiquitinated in position 353. The structural features of the conjugate were examined by circular dichroism and NMR spectroscopy. The ubiquitinated tau was challenged in a number of assays: fibrils formation under aggregating conditions in vitro, chemical stability upon exposure to a variety of biological media including cell extracts, and internalization into astrocytes. The results demonstrated the wide applicability of the new semisynthetic strategy for the investigation of ubiquitinated substrates in vitro or in cell, and in particular for studying if ubiquitination has a role in the molecular mechanisms that underlie the aberrant transition of tau into pathological aggregates

    A New Battle for History in the Twenty-First Century?

    Abstract This article engages with some of the questions raised by David Armitage and Jo Guldi’s “The Return of the longue durée: An Anglo-American Perspective” and their resonance among readers of the Annales. In particular, it challenges the authors’ classification of a variety of different historical studies of short periods of time under the rubric of “microhistory.” It also questions their argument that such studies are evidence of a “moral crisis” that supposedly dominated anglophone historiography from the cultural revolution of 1968 to the global financial crisis of 2008. Furthermore, the article contrasts the less conventional meanings that Fernand Braudel originally attributed to the longue durée with the ways that Armitage and Guldi interpret this expression. Finally, it asks how, in practice, historians are supposed to follow the authors’ invitation to move beyond specialized training and knowledge to produce sweeping new and original interpretations of millennia of human history.</jats:p
