    Nutritional Status Driving Infection by Trypanosoma cruzi: Lessons from Experimental Animals

    This paper reviews the scientific knowledge about protein-energy and micronutrient malnutrition in the context of Chagas disease, especially in experimental models. The search of articles was conducted using the electronic databases of SciELO (Scientific Electronic Library Online), PubMed and MEDLINE published between 1960 and March 2010. It was possible to verify that nutritional deficiencies (protein-energy malnutrition and micronutrient malnutrition) exert a direct effect on the infection by T. cruzi. However, little is known about the immunological mechanisms involved in the relationship “nutritional deficiencies and infection by T. cruzi”. A hundred years after the discovery of Chagas disease many aspects of this illness still require clarification, including the effects of nutritional deficiencies on immune and pathological mechanisms of T. cruzi infection

    Periódicos científicos brasileiros promovem a adesão de pesquisadores em doença de Chagas a princípios éticos internacionais?

    The ethical aspects of the Brazilian publications about human Chagas disease (CD) developed between 1996 and 2010 and the policy adopted by Brazilian medical journals were analyzed. Articles were selected on the SciELO Brazil data basis, and the evaluation of ethical aspects was based on the normative contents about ethics in research involving human experimentation according to the Brazilian resolution of the National Health Council no. 196/1996. The editorial policies of the section "Instructions to authors" were analyzed. In the period of 1996-2012, 58.9% of articles involving human Chagas disease did not refer to the fulfillment of the ethical aspects concerning research with human beings. In 80% of the journals, the requirements and confirmation of the information about ethical aspects in the studies of human CD were not observed. Although a failure in this type of service is still observed, awareness has been raised in federal agencies, educational institutions/research and publishing groups to standardize the procedures and ethical requirements for the Brazilian journals, reinforcing the fulfillment of the ethical parameters, according to the resolution of NHC no. 196/1996.Foram analisados os aspectos éticos das publicações brasileiras sobre a doença de Chagas humana desenvolvidas entre 1996 e 2010 e avaliadas as políticas editoriais dos periódicos médicos brasileiros sobre ética em pesquisa. Os artigos foram selecionados do banco de dados da SciELO Brasil. A avaliação dos aspectos éticos foi baseada nos conteúdos normativos sobre ética envolvendo pesquisa com seres humanos, conforme Resolução brasileira do Conselho Nacional de Saúde - CNS 196/1996. A política editorial dos periódicos foi analisada utilizando-se a seção "Instrução aos autores". No período de 1996-2012, 58,9% dos estudos envolvendo DC humana não informaram no texto sobre o cumprimento dos aspectos éticos, em relação à pesquisa com seres humanos. Em 80% dos periódicos avaliados o requerimento e confirmação das informações sobre os aspectos éticos nos estudos sobre a DC humana não foram observados. Apesar de haver ainda lacunas nesse tipo de citação conforme foi observado, tem-se verificado uma conscientização pelos órgãos federais, instituições de ensino/pesquisa e grupos editoriais para padronizar os procedimentos e orientações éticas por parte dos periódicos brasileiros, reforçando o cumprimento dos pressupostos éticos preconizados pela resolução CNS 196/1996

O objetivo é considerar o humor segundo mecanismos psicológicos para a produção e concepção de enunciados publicitários, utilizando-se para isso de uma abordagem metodológica baseada em pesquisa bibliográfica.     Chagas' disease: an update on immune mechanisms and therapeutic strategies

    The final decade of the 20th century was marked by an alarming resurgence in infectious diseases caused by tropical parasites belonging to the kinetoplastid protozoan order. Among the pathogenic trypanosomatids, some species are of particular interest due to their medical importance. These species include the agent responsible for Chagas' disease, Trypanosoma cruzi.     Immunological imbalance between IFN-³ and IL-10 levels in the sera of patients with the cardiac form of Chagas disease

    The immune response is crucial for protection against disease; however, immunological imbalances can lead to heart and digestive tract lesions in chagasic patients. Several studies have evaluated the cellular and humoral immune responses in chagasic patients in an attempt to correlate immunological findings with clinical forms of Chagas disease. Moreover, immunoglobulins and cytokines are important for parasitic control and are involved in lesion genesis. Here, cytokine and IgG isotype production were studied, using total epimastigote antigen on sera of chagasic patients with indeterminate (IND, n = 27) and cardiac (CARD, n = 16) forms of the disease. Samples from normal, uninfected individuals (NI, n = 30) were use as controls. The results showed that sera from both IND and CARD patients contained higher levels of Trypanosoma cruzi-specific IgG1 (IgG1) antibodies than sera from NI. No difference in IgG2 production levels was observed between NI, IND and CARD patients, nor was a difference in IL-10 and IFN-³ production detected in the sera of IND, CARD and NI patients. However, IND patients displayed a positive correlation between IL-10 and IFN-³ levels in serum, while CARD patients showed no such correlation, indicating an uncontrolled inflammatory response in CARD patients. These findings support the hypothesis that a lack of efficient regulation between IFN-³ and IL-10 productions in CARD patients may lead to cardiac immunopathology.CNP

    Role of the CX3CL1/CX3CR1 axis in iron metabolism and immune regulation during acute Trypanosoma cruzi infection

    IntroductionDuring Trypanosoma cruzi infection, the immune system activates a robust inflammatory response, involving cytokines and chemokines like IFN-γ, TNF, IL-6, IL-1β, CCL2, and CCL5, to control parasite replication. The CX3CL1 chemokine and its receptor, CX3CR1, have been implicated in amplifying inflammation through pathways like NF-κB, MAPKs, STATs, TLRs, and NLRs, contributing to tissue damage. This study evaluated the effects of blocking CX3CR1 with the allosteric antagonist AZD8797 in a murine model of acute T. cruzi infection. MethodsMale C57BL/6 mice were infected with 103 trypomastigote forms of T. cruzi (Y strain) and received AZD8797 (10 mg/kg) intraperitoneally for 10 days. On the 10th day, animals were euthanized and heart, skeletal muscle, and liver tissues were collected for CX3C L1 protein expression, biomarkers (IL-1β, IL-4, IL-6, IL-10, IL-15, IL-17, IFN-γ, TNF, and CCL2) quantified by Cytometric Bead Array and Enzyme Immunoassay. ResultsTreatment reduced spleen mass and cardiac levels of CCL2 and IL-15, with an increase of IL-4. Conversely, in skeletal muscle, TNF, IL-6, and IL-10 increased, while IL-15 decreased. Liver tissue showed reduced IL-15, IL-6, and IL-1β levels, alongside lowered plasma hepcidin and ferritin concentrations. DiscussionThese findings highlight CX3CL1’s site-specific role in modulating inflammation and iron metabolism during acute T. cruzi infection, suggesting its potential as a therapeutic target for infection management and disease prognosis
