Subtle balance of tropoelastin molecular shape and flexibility regulates dynamics and hierarchical assembly
The assembly of the tropoelastin monomer into elastin is vital for conferring elasticity on blood vessels, skin, and lungs. Tropoelastin has dual needs for flexibility and structure in self-assembly. We explore the structure-dynamics-function interplay, consider the duality of molecular order and disorder, and identify equally significant functional contributions by local and global structures. To study these organizational stratifications, we perturb a key hinge region by expressing an exon that is universally spliced out in human tropoelastins. We find a herniated nanostructure with a displaced C terminus and explain by molecular modeling that flexible helices are replaced with substantial β sheets. We see atypical higher-order cross-linking and inefficient assembly into discontinuous, thick elastic fibers. We explain this dysfunction by correlating local and global structural effects with changes in the molecule’s assembly dynamics. This work has general implications for our understanding of elastomeric proteins, which balance disordered regions with defined structural modules at multiple scales for functional assembly.United States. Unraveling the molecular mechanisms of thermo-responsive properties of silk-elastin-like proteins by integrating multiscale modeling and experiment
Adaptive hydrogels tailor-made from silk-elastin-like proteins (SELPs) possess excellent biocompatibility and biodegradability with properties that are tunable and responsive to multiple simultaneous external stimuli. To unravel the molecular mechanisms of their physical response to external stimuli in tandem with experiments, here we predict and measure the variation in structural properties as a function of temperature through coarse-grained (CG) modeling of individual and crosslinked SE8Y and S4E8Y molecules, which have ratios of 1:8 and 4:8 of silk to elastin blocks respectively. Extensive structural reshuffling in single SE8Y molecules led to the increased compactness of the structure, whereas S4E8Y molecules did not experience any significant changes as they already adopted very compact structures at low temperatures. Crosslinking of SE8Y molecules at high concentrations impeded their structural transition at high temperatures that drastically reduced the degree of deswelling through extensive suppression of the structural shuffling and the trapping of the molecules in high potential energy states due to inter-molecular constraints. This integrative experimental and computational understanding of the thermal response in single molecules of SELPs and their crosslinked networks should lead to further improvements in the properties of SELP hydrogels through predictive designs and their wider applications in biomaterials and tissue engineering.United States. In the article the basic directions of activity of the Russian Federation in the framework of the
Shanghai cooperation organization are considered. The conclusion is that Russia from the
moment of foundation of the Organization actively supports association of the member countries’ positions of the Shanghai cooperation organization in the sphere of ensuring regional security. The analysis was carried out of economic policy concerning Organization member countries. As a result revealed that the creation of energy club lobbied by the Russian Federation shows the paramount importance of the energy component of the countries cooperation of the Organization. Highlight some positions that determine the importance of Russian energy diplomacy in the activities of the Shanghai cooperation organization

PROVIDING REGIONAL SECURITY WITHIN THE FRAMEWORK OF THE SHANGHAI COOPERATION ORGANIZATION
События последних лет в странах Северной Африки и Ближнего Востока («арабская весна»), способствовали актуализации вопросов безопасности в деятельности региональных и глобальных организаций, в частности Шанхайской организации сотрудничества (ШОС). Основным направлением деятельности ШОС является укрепление региональной безопасности. Региональная антитеррористическая структура Шанхайской организации сотрудничества (РАТС ШОС) координирует деятельность, связанную с обменом информацией и укреплением сотрудничества в борьбе с терроризмом, экстремизмом и сепаратизмом.Целью исследования является определение эффективности работы РАТС ШОС в обеспечении региональной безопасности.При исследовании использовались методы как теоретического, так и эмпирического уровня. Особенно полезным оказался системный подход, который позволил рассмотреть деятельность внутренних структур РАТС ШОС и мероприятий проводимых в рамках данного института ШОС в совокупности.В результате выявлено, что ШОС постепенно становится важнейшим инструментом в борьбе с экстремизмом и терроризмом в центральноазиатском регионе. За последние два года, в рамках ШОС была проведена большая серия совместных антитеррористических, военных и военно-морских учений. Важно подчеркнуть, что разработанная договорно-правовая база Организации дает возможность осуществлять антитеррористические мероприятия в рамках международно-правового поля.Материалы статьи могут использоваться для сравнительного анализа работы различных структур, межгосударственных союзов, деятельность которых направлена на борьбу с региональным и международным терроризмом.Recent events in North Africa and the Middle East (the “Arab Spring”) contributed to the mainstreaming of security issues in the regional and global organizations, such as the Shanghai Cooperation Organization (SCO). The main activity of the SCO is a strengthen regional security. The Regional anti-terrorist structure of Shanghai Cooperation Organization (RATS SCO) coordinates activity connected with the information exchange and strengthening cooperation in the struggle against terrorism, extremism and separatism.The objective of the study is to determine the effectiveness of the RATS SCO in regional security.Methods of research are theoretical and empirical level. A systematic approach was particularly useful. He allowed to review the activities of the internal structures of the RATS SCO and activities in the framework of the SCO in as a whole.The results showed that SCO is gradually becoming an important tool in the struggle against extremism and terrorism in the Central Asia region. A large series of joint anti-terrorism, military and navy trainings were conducted in the framework of the SCO. It is significant that developed international legal base of the Organization makes it possible to carry out anti-terrorist activities in the international legal field.The results can be used for comparative analysis of the various interstate structures, whose activities are aimed at struggle with regional and international terrorism
