    This work, which follows on others, study the variation of the pH, and likewise the concentrations of the ions Ca (II) and SO<sub>4</sub> (II) of the filtered potable water in which there were submerged the test specimens of mortar (1:3) made with a Portland cement (cement 3 < > P-450) and with the mixtures cement 3/slag = 85/15 - 65/35 - 40/60 and 30/70 of weight, submerged for a curing period (21 days) and later during 56 - 90 - 180 and 360 days (conservation periods), in this stage. It also study the chemical composition of the new solid phases formed in the said means. The structural composition of the solid phases (calcite) was determined by DRX; an account of this was given in (2).
 In the different studied cases it has been demonstrated that an increase of the content of Ca (II) in the solution and in the new solid phases and of the value of the pH is produced, which depends on the mixture used to manufacture the different test specimen series and on the time of conservation. The content of Ca (II) in the solution is less than 0,1 X 10<sup>-2</sup> moles, in most of the cases, while in the new solid phases is greater, reaching 4,31 X 10<sup>-2</sup> moles.
 On the contrary the ion SO<sub>4</sub> (II) has not been in the solid phases mentioned, being found in little quantities in the different liquid phases (filtered potable water); the greater contents correspond to the means of the mixture that have the greater mounts of slag (60 and 70 % of weight).<br><br>En el presente trabajo, continuación de otros, se estudia la variación del pH, así como de la concentración de los iones Ca (II) y SO<sub>4</sub> (II) del agua potable filtrada en donde han estado sumergidas las series de probetas de mortero (1:3) hechas con un cemento portland (cemento 3 < > < > P-450) y con las mezclas cemento 3/escoria = 85/15 - 65/35 - 40/60 y 30/70, en peso, durante el período de curado (21 días) y, posteriormente, durante 56 - 90 - 180 y 360 días (períodos de conservación), en esta etapa. Así mismo, se estudia la composición química de las nuevas fases sólidas formadas en dichos medios. La composición estructural de dichas fases sólidas (calcita) se determinó por DRX, de ello se dio cuenta en (2).
 En los distintos casos estudiados se ha puesto de manifiesto que se produce un incremento del contenido de Ca (II) en la disolución y en las nuevas fases sólidas, así como del valor del pH, que dependen de la mezcla utilizada para fabricar las diversas series de probetas y del tiempo de conservación. Las cantidades de Ca (II) en la disolución son, en la mayoría de los casos menores de 0,1 X 10<sup>-2</sup> moles, mientras que en las nuevas fases sólidas son mayores, llegando a alcanzar valores de 4,31 X X 10<sup>-2</sup> moles.
 Por el contrario, el ion SO<sub>4</sub> (II) no se ha detectado en las mencionadas fases sólidas, encontrándose en pequeñas cantidades en las diversas fases líquidas (agua potable filtrada); los mayores contenidos corresponden a los medios de las mezclas que tienen las mayores cantidades de escoria (60 y 70 %, en peso)

    Introducing automation to the molecular diagnosis of Trypanosoma cruzi infection: A comparative study of sample treatments, DNA extraction methods and real-time PCR assays

    Background Polymerase chain reaction (PCR) has become a useful tool for the diagnosis of Trypanosoma cruzi infection. The development of automated DNA extraction methodologies and PCR systems is an important step toward the standardization of protocols in routine diagnosis. To date, there are only two commercially available Real-Time PCR assays for the routine laboratory detection of T. cruzi DNA in clinical samples: TCRUZIDNA.CE (Diagnostic Bioprobes Srl) and RealCycler CHAG (Progenie Molecular). Our aim was to evaluate the RealCycler CHAG assay taking into account the whole process. Methodology/Principal findings We assessed the usefulness of an automated DNA extraction system based on magnetic particles (EZ1 Virus Mini Kit v2.0, Qiagen) combined with a commercially available Real-Time PCR assay targeting satellite DNA (SatDNA) of T. cruzi (RealCycler CHAG), a methodology used for routine diagnosis in our hospital. It was compared with a well-known strategy combining a commercial DNA isolation kit based on silica columns (High Pure PCR Template Preparation Kit, Roche Diagnostics) with an in-house Real-Time PCR targeting SatDNA. The results of the two methodologies were in almost perfect agreement, indicating they can be used interchangeably. However, when variations in protocol factors were applied (sample treatment, extraction method and Real-Time PCR), the results were less convincing. A comprehensive fine-tuning of the whole procedure is the key to successful results. Guanidine EDTA-blood (GEB) samples are not suitable for DNA extraction based on magnetic particles due to inhibition, at least when samples are not processed immediately. Conclusions/Significance This is the first study to evaluate the RealCycler CHAG assay taking into account the overall process, including three variables (sample treatment, extraction method and Real-Time PCR). Our findings may contribute to the harmonization of protocols between laboratories and to a wider application of Real-Time PCR in molecular diagnostic laboratories associated with health centers.Fil: Abras, Alba.     Epidermal growth factor secreted from submandibular salivary glands interferes with the lipolytic effect of adrenaline in mice

    We had described that epidermal growth factor (EGF) interfered with the lipolytic effect of catecholamines in isolated adipocytes. Since catecholamines stimulate the release of EGF from submandibular salivary glands to blood plasma in male mice, we studied whether EGF affected also the lipolytic response to adrenaline in whole animals. We studied the effect of adrenaline in sialoadenectomized and sham-operated mice receiving or not a high dose of EGF following adrenaline injection. There was no difference in plasma EGF concentration between sham-operated and sialoadenectomized animals receiving saline. After adrenaline administration plasma EGF increased by 20-fold in sham-operated but did not increase in sialoadenectomized mice. Indeed, the increase was much higher (more than 100-fold) in mice receiving exogenous EGF. The effect of adrenaline on plasma concentration of both glycerol and nonesterified fatty acids was higher as lower was plasma EGF concentration. Isolated adipocytes obtained from sham-operated or sialoadenectomized mice had identical lipolytic response to adrenaline. The lipolytic response of adipocytes to isoproterenol was decreased by addition of EGF. To study whether the interference with the in vivo lipolytic effect of adrenaline had further metabolic consequences, we measured plasma b-hydroxybutyrate concentration in plasma. There was no difference in the response to adrenaline between sham-operated and sialoadenectomized mice in spite of the difference in plasma nonsterified fatty acid concentration. Studies in isolated hepatocytes indicated that ketogenesis run at near maximal rate in this range of substrate concentration. These results suggest that EGF in the physiological range decreases the lipolytic effect of adrenaline but does not compromise further metabolic events like the enhancement of ketogenesis

    Oncogenic K-Ras segregates at spatially distinct plasma membrane signaling platforms according to its phosphorylation status

    Activating mutations in the K-Ras small GTPase are extensively found in human tumors. Although these mutations induce the generation of a constitutively GTP-loaded, active form of K-Ras, phosphorylation at Ser181 within the C-terminal hypervariable region can modulate oncogenic K-Ras function without affecting the in vitro affinity for its effector Raf-1. In striking contrast, K-Ras phosphorylated at Ser181 shows increased interaction in cells with the active form of Raf-1 and with p110α, the catalytic subunit of PI 3-kinase. Because the majority of phosphorylated K-Ras is located at the plasma membrane, different localization within this membrane according to the phosphorylation status was explored. Density-gradient fractionation of the plasma membrane in the absence of detergents showed segregation of K-Ras mutants that carry a phosphomimetic or unphosphorylatable serine residue (S181D or S181A, respectively). Moreover, statistical analysis of immunoelectron microscopy showed that both phosphorylation mutants form distinct nanoclusters that do not overlap. Finally, induction of oncogenic K-Ras phosphorylation - by activation of protein kinase C (PKC) - increased its co-clustering with the phosphomimetic K-Ras mutant, whereas (when PKC is inhibited) non-phosphorylated oncogenic K-Ras clusters with the non-phosphorylatable K-Ras mutant. Most interestingly, PI 3-kinase (p110α) was found in phosphorylated K-Ras nanoclusters but not in non-phosphorylated K-Ras nanoclusters. In conclusion, our data provide - for the first time - evidence that PKC-dependent phosphorylation of oncogenic K-Ras induced its segregation in spatially distinct nanoclusters at the plasma membrane that, in turn, favor activation of Raf-1 and PI 3-kinase

    Temporal changes in inhibin subunit mRNAs during atresia of preovulatory follicles in the rat

    This study aimed to investigate the time course of disappearance of the mRNAs of the various subunits of inhibin in follicles which become atretic. An animal model was used in which atresia of preovulatory follicles could be studied in a chronological order. Injection of gonadotrophin-releasing hormone (GnRH) antagonist (20 microg) at the morning of pro-oestrus (P) blocked ovulation and the 10-12 preovulatory follicles became gradually atretic. A second injection was given the next day to prevent delayed ovulation. The rate of atresia could be delayed by simultaneous administration of a subovulatory dose of human chorionic gonadotrophin (hCG) (0.5 IU) and could be advanced by administration of a fivefold larger amount of GnRH antagonist. Functional activity of follicles becoming atretic was studied by measuring oestradiol production after incubation of individual follicles for 4 h. Follicles isolated 24 h after the first injection of GnRH antagonist (P+24) already secreted significantly less oestradiol in vitro than follicles isolated at pro-oestrus, although they were morphologically not different from pro-oestrous follicles. Follicles isolated at P+24 from hCG-treated rats secreted more oestradiol compared with follicles from rats not treated with hCG. In contrast, follicles isolated at P+24 from rats that were given a fivefold larger amount of GnRH antagonist secreted less oestradiol. Once this model was validated, temporal changes in inhibin subunit mRNAs in follicles undergoing atresia were measured by in situ hybridization and RNase protection assay. In situ hybridization showed abundant alpha- and betaA-subunit mRNA in the whole granulosa layer of preovulatory follicles at P and P+24, while betaB-subunit mRNA was restricted to the antral layer and cumulus. At P+48 the amount of alpha- and betaA-subunit mRNA had declined and was restricted to the cumulus, whereas betaB-subunit mRNA was absent. In the atretic follicles present at P+72 and P+96, mRNAs of all three inhibin subunits were absent. Administration of 0.5 IU hCG delayed the decline in the amount of alpha, betaA and betaB mRNA in preovulatory follicles at P+48.     Colorimetric and spectral analysis of rock art by means of the characterization of digital sensors

    Tesis por compendio[ES] Las labores de documentación de arte rupestre son arduas y delicadas, donde el color desempeña un papel fundamental, proporcionando información vital a nivel descriptivo, técnico y cuantitativo . Tradicionalmente los métodos de documentación en arqueología quedaban restringidos a procedimientos estrictamente subjetivos. Sin embargo, esta metodología conlleva limitaciones prácticas y técnicas, afectando a los resultados obtenidos en la determinación del color. El empleo combinado de técnicas geomáticas, como la fotogrametría o el láser escáner, junto con técnicas de procesamiento de imágenes digitales, ha supuesto un notable avance. El problema es que, aunque las imágenes digitales permiten capturar el color de forma rápida, sencilla, y no invasiva, los datos RGB registrados por la cámara no tienen un sentido colorimétrico riguroso. Se requiere la aplicación de un proceso riguroso de tranformación que permita obtener datos fidedignos del color a través de imágenes digitales. En esta tesis se propone una solución científica novedosa y de vanguardia, en la que se persigue integrar el análisis espectrofotométrico y colorimétrico como complemento a técnicas fotogramétricas que permitan una mejora en la identificación del color y representación de pigmentos con máxima fiabilidad en levantamientos, modelos y reconstrucciones tridimensionales (3D). La metodología propuesta se basa en la caracterización colorimétrica de sensores digitales, que es de novel aplicación en pinturas rupestres. La caracterización pretende obtener las ecuaciones de transformación entre los datos de color registrados por la cámara, dependientes del dispositivo, y espacios de color independientes, de base física, como los establecidos por la Commission Internationale de l'Éclairage (CIE). Para el tratamiento de datos colorimétricos y espectrales se requiere disponer de un software de características técnicas muy específicas. Aunque existe software comercial, lo cierto es que realizan por separado el tratamiento digital de imágenes y las operaciones colorimétricas. No existe software que integre ambas, ni que además permita llevar a cabo la caracterización. Como aspecto fundamental, presentamos en esta tesis el software propio desarrollado, denominado pyColourimetry, siguiendo las recomendaciones publicadas por la CIE, de código abierto, y adaptado al flujo metodológico propuesto, de modo que facilite la independencia y el progreso científico sin ataduras comerciales, permitiendo el tratamiento de datos colorimétricos y espectrales, y confiriendo al usuario pleno control del proceso y la gestión de los datos obtenidos. Adicinalmente, en este estudio se expone el análisis de los principales factores que afectan a la caracterización tales como el sensor empleado, los parámetros de la cámara durante la toma, la iluminación, el modelo de regresión, y el conjunto de datos empleados como entrenamiento del modelo. Se ha aplicado un modelo de regresión basado en procesos Gaussianos, y se ha comparado con los resultados obtenidos mediante polinomios. También presentamos un nuevo esquema de trabajo que permite la selección automática de muestras de color, adaptado al rango cromático de la escena, que se ha denominado P-ASK, basado en el algoritmo de clasificación K-means. Los resultados obtenidos en esta tesis demuestran que el proceso metodológico de caracterización propuesto es altamente aplicable en tareas de documentación y preservación del patrimonio cultural en general, y en arte rupestre en particular. Se trata de una metodología de bajo coste, no invasiva, que permite obtener el registro colorimétrico de escenas completas. Una vez caracterizada, una cámara digital convencional puede emplearse para la determinación del color de forma rigurosa, simulando un colorímetro, lo que permitirá trabajar en un espacio de color de base física, independiente del dispositivo y comparable con[CA] Les tasques de documentació gràfica d'art rupestre són àrdues i delicades, on el color compleix un paper fonamental, proporcionant informació vital a nivell descriptiu, t\`ecnic i quantitatiu.Tradicionalment els mètodes de documentació en arqueologia quedaven restringits a procediments estrictament subjectius, comportant limitacions pràctiques i tècniques, afectant els resultats obtinguts en la determinació de la color. Es requereix l'aplicació d'un procés rigorós de transformació que permeti obtenir dades fidedignes de la color a través d'imatges digitals. En aquesta tesi es proposa una solució científica innovadora i d'avantguarda, en la qual es persegueix integrar l'anàlisi espectrofotomètric i colorimètric com a complement a tècniques fotogramètriques que permetin una millora en la identificació de la color i representació de pigments amb màxima fiabilitat en aixecaments, models i reconstruccions tridimensionals 3D. La metodologia proposada es basa en la caracterització colorimètrica de sensors digitals, que és de novell aplicació en pintures rupestres. La caracterització pretén obtenir les equacions de transformació entre les dades de color registrats per la càmera, dependents d'el dispositiu, i espais de color independents, de base física, com els establerts per la Commission Internationale de l'Éclairage (CIE). Per al tractament de dades colorimètriques i espectrals de forma rigorosa es requereix disposar d'un programari de característiques tècniques molt específiques. Encara que hi ha programari comercial, fan per separat el tractament digital d'imatges i les operacions colorimètriques. No hi ha programari que integri totes dues, ni que permeti dur a terme la caracterització. Com a aspecte addicional i fonamental, vam presentar el programari propi que s'ha desenvolupat, denominat pyColourimetry, segons les recomanacions publicades per la CIE, de codi obert, i adaptat al flux metodológic proposat, de manera que faciliti la independència i el progrés científic sense lligams comercials, permetent el tractament de dades colorimètriques i espectrals, i conferint a l'usuari ple control del procés i la gestió de les dades obtingudes. A més, s'exposa l'anàlisi dels principals factors que afecten la caracterització tals com el sensor emprat, els paràmetres de la càmera durant la presa, il¿luminació, el model de regressió, i el conjunt de dades emprades com a entrenament d'el model. S'ha aplicat un model de regressió basat en processos Gaussians, i s'han comparat els resultats obtinguts mitjançant polinomis. També vam presentar un nou esquema de treball que permet la selecció automàtica de mostres de color, adaptat a la franja cromàtica de l'escena, que s'ha anomenat P-ASK, basat en l'algoritme de classificació K-means. Els resultats obtinguts en aquesta tesi demostren que el procés metodològic de caracterització proposat és altament aplicable en tasques de documentació i preservació de el patrimoni cultural en general, i en art rupestre en particular. Es tracta d'una metodologia de baix cost, no invasiva, que permet obtenir el registre colorimètric d'escenes completes. Un cop caracteritzada, una càmera digital convencional pot emprar-se per a la determinació de la color de forma rigorosa, simulant un colorímetre, el que permetrà treballar en un espai de color de base física, independent d'el dispositiu i comparable amb dades obtingudes mitjançant altres càmeres que tambè estiguin caracteritzades.[EN] Cultural heritage documentation and preservation is an arduous and delicate task in which color plays a fundamental role. The correct determination of color provides vital information on a descriptive, technical and quantitative level. Classical color documentation methods in archaeology were usually restricted to strictly subjective procedures. However, this methodology has practical and technical limitations, affecting the results obtained in the determination of color. Nowadays, it is frequent to support classical methods with geomatics techniques, such as photogrammetry or laser scanning, together with digital image processing. Although digital images allow color to be captured quickly, easily, and in a non-invasive way, the RGB data provided by the camera does not itself have a rigorous colorimetric sense. Therefore, a rigorous transformation process to obtain reliable color data from digital images is required. This thesis proposes a novel technical solution, in which the integration of spectrophotometric and colorimetric analysis is intended as a complement to photogrammetric techniques that allow an improvement in color identification and representation of pigments with maximum reliability in 3D surveys, models and reconstructions. The proposed methodology is based on the colorimetric characterization of digital sensors, which is of novel application in cave paintings. The characterization aims to obtain the transformation equations between the device-dependent color data recorded by the camera and the independent, physically-based color spaces, such as those established by the Commission Internationale de l'Éclairage (CIE). The rigorous processing of color and spectral data requires software packages with specific colorimetric functionalities. Although there are different commercial software options, they do not integrate the digital image processing and colorimetric computations together. And more importantly, they do not allow the camera characterization to be carried out. Therefore, as a key aspect in this thesis is our in-house pyColourimetry software that was developed and tested taking into account the recommendations published by the CIE. pyColourimetry is an open-source code, independent without commercial ties; it allows the treatment of colorimetric and spectral data and the digital image processing, and gives full control of the characterization process and the management of the obtained data to the user. On the other hand, this study presents a further analysis of the main factors affecting the characterization, such as the camera built-in sensor, the camera parameters, the illuminant, the regression model, and the data set used for model training. For computing the transformation equations, the literature recommends the use of polynomial equations as a regression model. Thus, polynomial models are considered as a starting point in this thesis. Additionally, a regression model based on Gaussian processes has been applied, and the results obtained by means of polynomials have been compared. Also, a new working scheme was reported which allows the automatic selection of color samples, adapted to the chromatic range of the scene. This scheme is called P-ASK, based on the K-means classification algorithm. The results achieved in this thesis show that the proposed framework for camera characterization is highly applicable in documentation and conservation tasks in general cultural heritage applications, and particularly in rock art painting. It is a low-cost and non-invasive methodology that allows for the colorimetric recording from complete image scenes. Once characterized, a conventional digital camera can be used for rigorous color determination, simulating a colorimeter.
