Accessible Internet-of-Things and Internet-of-Content Services for All in the Home or on the Move
Public authorities and national healthcare service providers across the European Union and beyond are increasingly under pressure to deliver better services with diminishing resources. Users, quite rightly, demand a high standard of service and increasingly also convenience focused on the needs of the users rather than how the services are organised to deliver them. Users, on the whole, also prefer to be supported in their homes, and be free to move as much as possible. While users without impairments and generally well provided for, there are increasingly also users who have special needs, including the aged and those with a range of physical and mental disabilities who are unable to make effective use of such services from their homes due to the lack of proactive and accessible services.
The idea we promote in the chapter is to provide better support to users with disabilities and impairments from the comfort of their home by means of providing them with a set of scalable services which can be either offered for free or purchased through some central form of a marketplace repository
Non-rigid multi-frame registration of cell nuclei in live cell microscopy image data
To gain a better understanding of cellular and molecular processes it is important to quantitatively analyze the motion of subcellular particles in live cell microscopy image sequences. For accurate quantification of the subcellular particle motion, compensation of the motion and deformation of the cell nucleus is required. This thesis deals with non-rigid registration of cell nuclei in 2D and 3D live cell fluorescence microscopy images. We developed two multi-frame non-rigid registration approaches which simultaneously exploit information from multiple consecutive frames of an image sequence to improve the registration accuracy. The multi-frame registration approaches are based on local optic flow estimation, use information from multiple consecutive images, and
take into account computed transformations from previous time steps. The first approach comprises three intensity-based variants and two different temporal weighting schemes. The second approach determines diffeomorphic transformations in the log-domain which allows efficient computation of the inverse transformations. We use a temporally weighted mean image which is constructed based on inverse transformations and multiple consecutive frames. In addition, we employ a flow boundary preserving method for regularization of computed deformation vector fields. Both multi-frame registration approaches have been successfully applied to 2D and 3D synthetic as well as real live cell microscopy image sequences. We have performed an extensive quantitative evaluation of our approaches and compared their performance with previous non-rigid pairwise, multi-frame, and temporal groupwise registration approaches
The Role of Diet in the Management of Psoriasis: A Scoping Review

Globally, there are an estimated 60 million people living with psoriasis (PLwP). There is a growing body of evidence on the role of diet in psoriasis management and demand for dietary advice is high. However, there are no specific, evidence-based dietary guidelines. This scoping review summarises the literature on use and effectiveness of diet in the management of psoriasis to improve understanding of the evidence and assist PLwP and healthcare professionals (HCPs) to discuss diet. The findings were categorised into three themes (1) dietary intakes of PLwP, (2) the perceived role of diet in psoriasis management and (3) dietary approaches to manage psoriasis symptoms. In cross-sectional studies PLwP were reported to have higher fat and lower fibre intakes compared to controls, and lower psoriasis severity was associated with higher fibre intake. However, research is limited. PLwP perceive diet to have an impact on symptoms and make dietary modifications which are often restrictive. Systematic reviews and RCTs found certain dietary approaches improved symptoms, but only in specific populations (e.g., PLwP with obesity and PLwP with coeliac disease), and evidence for supplement use is inconclusive. The grey literature provides limited guidance to PLwP; focusing on weight-loss and associated comorbidities. Larger, controlled trials are required to determine dietary approaches for psoriasis management, especially in PLwP without obesity and non-coeliac PLwP. Further understanding of diet modification, information acquisition and experiences among PLwP will enhance holistic care for psoriasis management.Peer reviewe
Qualitative exploration of the experiences and perceptions of diet in psoriasis management among UK adults
Objective: This study aimed to explore the use, experiences and perceptions of diet in psoriasis management among adults with lived experience in the UK. Design: Qualitative. Data were analysed thematically using a reflexive thematic approach. Setting: Online discussions with adults living with psoriasis in the UK. Participants: Nine adults (two men, seven women) ≥18 years of age, living in the UK, English speaking, with a diagnosis of psoriasis of any severity. Results: Four key themes were generated: (1) impact of diet, (2) dietary modification, (3) dietary information and (4) dietary support. Overall, the majority (n=8) perceived that diet had an impact on their psoriasis. Most participants (n=7) reported trying restrictive diets including dairy free, gluten free and ‘cleanses’ to help manage their psoriasis with limited success. A perceived lack of dietary support resulted in participants relying on social media and online forums for dietary information. Participants reported a high cognitive burden due to the lack of reliable nutrition guidance and insufficient dietary support from healthcare professionals (HCPs). Conclusions: Participants rely on social media and online forums for dietary information, which suggest unsubstantiated restrictive diets that could negatively impact health. Participants felt overwhelmed by dietary recommendations and wanted more relevant dietary support. In the absence of evidence-based dietary information for psoriasis, HCPs need to be able to provide basic dietary support and combat misinformation. Larger studies aimed at understanding how best to support people with psoriasis are needed
Diet quality in late midlife is associated with faster walking speed in later life in women, but not men: findings from a prospective British birth cohort
Healthy diet has been linked to better age-related physical functioning, but evidence on the relationship of overall diet quality in late midlife and clinically relevant measures of physical functioning in later life is limited. Research on potential sex differences in this relationship is scarce. The aim was to investigate the prospective association between overall diet quality, as assessed by the Healthy Eating Index-2015 at age 60-64y and measures of walking speed seven years later, among men and women from the Insight46, a neuroscience sub-study of the Medical Research Council National Survey of Health and Development. Diet was assessed at age 60-64y using five-day food diaries, from which total HEI-2015 was calculated. At age 69-71y, walking speed was estimated during four 10-meter walks at self-selected pace, using inertial measurement units. Multivariable linear regression models with sex as modifier, controlling for age, follow-up, lifestyle, health, social variables and physical performance were used. The final sample was 164 women and 167 men (n=331). Women had higher HEI-2015 scores and slower walking speed than men. A 10 point increase in HEI-2015 was associated with faster walking speed seven years later among women (B: 0.024, 95% CI: 0.006, 0.043), but not men. The association remained significant in the multivariable model (B: 0.021, 95% CI: 0.003, 0.040). In women in late midlife higher diet quality is associated with faster walking speed. A healthy diet in late midlife is likely to contribute towards better age-related physical capability and sex differences are likely to affect this relationship

Non-rigid Contour-Based Registration of Cell Nuclei in 2D Live Cell Microscopy Images Using a Dynamic Elasticity Model
International audienceThe analysis of the pure motion of subnuclear structures without influence of the cell nucleus motion and deformation is essential in live cell imaging. In this work, we propose a 2D contour-based image registration approach for compensation of nucleus motion and deformation in fluorescence microscopy time-lapse sequences. The proposed approach extends our previous approach which uses a static elasticity model to register cell images. Compared to that scheme, the new approach employs a dynamic elasticity model for forward simulation of nucleus motion and deformation based on the motion of its contours. The contour matching process is embedded as a constraint into the system of equations describing the elastic behavior of the nucleus. This results in better performance in terms of the registration accuracy. Our approach was successfully applied to real live cell microscopy image sequences of different types of cells including image data that was specifically designed and acquired for evaluation of cell image registration methods. An experimental comparison with existing contour-based registration methods and an intensity-based registration method has been performed. We also studied the dependence of the results on the choice of method parameters

Mediterranean and MIND dietary patterns and cognitive performance in multiple sclerosis: A cross-sectional analysis of the UK Multiple Sclerosis Register
Background: Multiple sclerosis (MS) is a chronic auto-immune neuroinflammatory disorder presenting as a range of systemic and neurological symptoms, including cognitive impairment. Emerging evidence suggests that diets targeting brain health—such as the Mediterranean (MED) and Mediterranean-DASH Intervention for Neurodegenerative Delay (MIND) diets—may improve cognitive function; however, studies examining their role in people living with MS are limited. Methods: We examined cross-sectional associations between diet and cognition data from 967 participants in the United Kingdom Multiple Sclerosis Register (UKMSR). Dietary pattern scores (alternate Mediterranean; aMED, and MIND) were derived from the 130-item EPIC-Norfolk food frequency questionnaire. Cognition was assessed using the MS-specific Cognitron-MS (C-MS) battery (13 tasks) and summarised as overall cognition (global G factor) and four domains (object memory, problem solving, information processing speed [IPS], and words memory). Cognitive outcomes were expressed as Deviation-from-Expected (DfE) scores standardised to demographic and device characteristics using external regression-based norms. Linear models were adjusted for total energy intake, MS phenotype, disease duration since diagnosis, and current disease-modifying therapy (DMT) use. Interactions tested moderation by MS phenotype (relapsing vs. progressive MS) and current DMT use (yes vs. no). Sensitivity analyses included within-domain multiple-comparison control, rank-based inverse-normal transformation, and winsorisation. Results: Greater alignment with aMED and MIND dietary patterns were associated with higher scores in specific cognitive domains but not in overall cognition. Higher aMED scores were associated most consistently with better IPS, while higher MIND scores were additionally associated with better words memory. In categorical models, participants in the middle or highest tertiles of aMED or MIND scores performed up to ~0.4 SD better on tasks of Verbal Analogies, Word Definitions, Simple Reaction Time, Words Memory Immediate, or Words Memory Delays compared with those in the lowest tertile. These findings were robust across sensitivity analyses. Stratified analyses showed differential cognitive performance and diet-cognition associations by MS phenotype and DMT use. Conclusions: Mediterranean and MIND dietary patterns showed modest cross-sectional associations with specific cognition domains, with differential cognitive performance in different subgroups according to MS phenotype and DMT use. Although causal inference is not possible, our findings indicate future MS-related dietary studies (longitudinal and/or randomised controlled trials) examining cognitive function domains across different MS subgroups are warranted
