    Interpolation problems in meteor radar analysis, part 7.6A

    Meteor echoes come from random points in the observation volume, and are irregularly spaced in time. This precludes the use of fast fourier transformations (FFT) techniques on the raw data to give the spectrum of waves that are present. One way around this obstacle is to restrict our interest to a particular class of waves, and fit a corresponding model to the raw data. It is assumed that there is no horizontal variation across the observation volume for tides, but in the vertical this is certainly not the case. If, in addition, we are interested in other types of waves which may be present and whose periods are unknown, then examining the raw line-of-sight velocities does not tell us how to modify the model, since the line-of-sight direction is not fixed. This is the motivation for interpolation. Interpolation takes a temporal series of line-of-sight velocities, and transforms it to a temporal series of wind velocities for each orthogonal direction. The velocities along a given direction can then be examined readily for any waves in addition to tides

    Biochemical Analysis of the Protein-Protein Interactions Involved in Karyopherin-Mediated Transport Across the Nuclear Pore Complex

    Nucleocytoplasmic transport occurs through the nuclear pore complex (NPC), which in yeast is a highly symmetric ~50 MDa complex consisting of approximately 30 different proteins. Small molecules can freely exchange through the NPC, but macromolecules larger than ~40 kDa such as proteins, mRNAs, and ribosomal subunits must be aided across by shuttle proteins (karyopherins, or Kaps). Kap-mediated transport involves FG-nups, a family of NPC proteins. While much has been learned about the mechanism of nucleocytoplasmic transport, many details are still unknown; perhaps among the most important missing details is the binding kinetics of almost all the transport relevant interactions, due to significant technical challenges. The aim of this work is to analyze the protein-protein interactions involved in Kap-mediated transport across the NPC, using biochemical, biophysical, and cell biological approaches. Yeast karyopherins, model cargoes, and full-length FG-nups are enriched from bacteria, and their affinities are studied quantitatively. The presence of competitor proteins and changes in bait protein distribution are seen to effect apparent affinity of these interactions. The relevance of the in vitro Kap/NLS-cargo binding measurements is confirmed with a nucleocytoplasmic import assay that allows quantitative measurements of import to be made within single living cells. Trends observed in vitro for Kap/FG-nup interactions were consistent with ex vivo observations of interactions of transport factors with Xenopus oocyte NPCs and also with in vitro measurements of transport through a synthetic NPC-based filter. This work has suggested a role for factors such as non-specific competition in determining the kinetics and selectivity of transport

    Family Predictors of Quality of Life and Child Problem Behavior in Families of Young Children with Autism Spectrum Disorders

    Previous research on young children with Autism Spectrum Disorders (ASD) has primarily focused on the affected child, while often overlooking the family of the child with ASD. Multiple child-related variables have been linked with quality of life and problem behavior; however the role of family variables has not yet been evaluated in a systematic way. The purpose of the present study was to evaluate the association between (a) variables within the family system (i.e. relationship discord, social isolation, parental depression, and elevated parental stress) and (b) quality of life and problem behavior. One hundred and one mothers of 2-8 year old children with ASD participated. Assessment of problem behavior, quality of life, maternal depression, relationship discord, parental stress, and social support was conducted. Bivariate correlations and multiple regression analyses were used to evaluate the association among these variables. Relationship discord, maternal depression, and parental stress were associated with (a) high levels of problem behavior in young children with ASDs and (b) poor quality of life for both the child and the family. The role of the family in ASDs and implications for future intervention research is discussed

    Nanoscale stiffness topography reveals structure and mechanics of the transport barrier in intact nuclear pore complexes

    The nuclear pore complex (NPC) is the gate for transport between the cell nucleus and the cytoplasm. Small molecules cross the NPC by passive diffusion, but molecules larger than ∼5 nm must bind to nuclear transport receptors to overcome a selective barrier within the NPC1. Although the structure and shape of the cytoplasmic ring of the NPC are relatively well characterized2, 3, 4, 5, the selective barrier is situated deep within the central channel of the NPC and depends critically on unstructured nuclear pore proteins5, 6, and is therefore not well understood. Here, we show that stiffness topography7 with sharp atomic force microscopy tips can generate nanoscale cross-sections of the NPC. The cross-sections reveal two distinct structures, a cytoplasmic ring and a central plug structure, which are consistent with the three-dimensional NPC structure derived from electron microscopy2, 3, 4, 5. The central plug persists after reactivation of the transport cycle and resultant cargo release, indicating that the plug is an intrinsic part of the NPC barrier. Added nuclear transport receptors accumulate on the intact transport barrier and lead to a homogenization of the barrier stiffness. The observed nanomechanical properties in the NPC indicate the presence of a cohesive barrier to transport and are quantitatively consistent with the presence of a central condensate of nuclear pore proteins in the NPC channel

    Is Speed of Integration really a Success Factor of Mergers and Acquisitions? : An Analysis of the Role of Internal and External Relatedness

    Previous research on mergers and acquisitions (M&A) has neglected the issue of speed of post merger integration (PMI) by and large. This paper argues that there are benefits and detriments associated with speed of integration. Thus, in some situations speed may be highly beneficial whereas in others it may be harmful to the success of a merger or acquisition. It is argued that the benefits and detriments of speed of integration depend on the magnitude of internal and external relatedness between the merging firms prior to the merger or acquisition. Results from a survey of 232 horizontal mergers and acquisitions show that speed is most beneficial when external relatedness is low and at the same time internal relatedness is high. In contrast, speed is highly detrimental in the case of low internal and high external relatedness

    Big data in an HR context: Exploring organizational change readiness, employee attitudes and behaviors

    YesThis research highlights a contextual application for big data within a HR case study setting. This is achieved through the development of a normative conceptual model that seeks to envelop employee behaviors and attitudes in the context of organizational change readiness. This empirical application considers a data sample from a large public sector organization and through applying Structural Equation Modelling (SEM) identifies salary, job promotion, organizational loyalty and organizational identity influences on employee job satisfaction (suggesting and mediating employee readiness for organizational change). However in considering this specific context, the authors highlight how, where and why such a normative approach to employee factors may be limited and thus, proposes through a framework which brings together big data principles, implementation approaches and management commitment requirements can be applied and harnessed more effectively in order to assess employee attitudes and behaviors as part of wider HR predictive analytics (HRPA) approaches. The researchers conclude with a discussion on these research elements and a set of practical, conceptual and management implications of the findings along with recommendations for future research in the area
