    Dissecting the Entry Pathways of Hepatitis C and Bovine Viral Diarrhea Viruses

    Hepatitis C virus (HCV) and bovine viral diarrhea virus (BVDV) are closely related members of the family Flaviviridae and are important human and animal pathogens, respectively. In this work, I investigated how these viruses interact with and enter both naïve and previously infected cells, specifically, the mechanisms of superinfection exclusion, the phenomenon by which previous viral infection prevents reinfection of the same cell, and the pathways of entry into target cells for these viruses. BVDV-acutely infected cells establish two blocks to superinfection, at the levels of virus entry and RNA replication. The former is mediated by the BVDV E2 protein, which was also identified as a transdominant inhibitor of BVDV entry in a screen of a random fragment BVDV cDNA library. The inhibitory region was further mapped to the E2 ectodomain and the block to entry was shown to occur downstream of CD46 receptor expression and BVDV binding, suggesting interference with a yet unidentified BVDV entry factor. In contrast to BVDV, HCV-infected and replicon-containing cells exhibit a post-entry block to superinfection, at one or more steps in viral replication. The entry pathways of both BVDV and HCV into target cells were also examined and found to be pH-dependent. In addition, these viruses were shown to be very acid-resistant, suggesting that they require an activation step to trigger pH-dependent entry. The role of the cellular receptors/coreceptors, CD81, scavenger receptor class B type 1 (SR-BI), and claudin-1 (CLDN1), in mediating HCV entry and their modulation during persistent HCV replication were also investigated. This work highlights common features shared between BVDV and HCV and provides insight into the cellular environment required for productive virus entry and RNA replication

    What Would Mr. Rogers Do? Healing Childhood Trauma Through the Application of Emergency Pedagogy

    Emergency Pedagogy works to stabilize, both physically and socially, those that have been affected by traumatic events through pedagogical interventions that are based on the concepts of Waldorf education (Ruf, 2013). While techniques and interventions such as Trauma-Informed Care and psychotherapy are more widely known and used to treat individuals who have experienced traumatic events, Emergency Pedagogy is another method to be considered when working with traumatized individuals (Cohen and Mannarino, 2016). Emergency Pedagogy is not therapy but rather pedagogical first aid, seeking to stabilize, both physically and socially, those that have been affected by traumatic events before the wound worsens and becomes post-traumatic stress disorder (Ruf, 2013). This dissertation will consider the works of educational theorists like Maslow and Bloom, as well as philosopher Nussbaum to gain a better understanding of the needs that must be met for a person to live a life of meaning, followed by the work of Ruf and Rogers whose focus is on the emotional well-being of children and helping them work through situations that are challenging to understand. This dissertation will also consider the work done by Oberman to bring Emergency Pedagogy into a public Waldorf charter school setting. Through interviews with key individuals this dissertation seeks to determine how to best bring “an expression of care” into non-Waldorf school settings and spaces through the application of Emergency Pedagogy in order to help children work through traumatic events.     Motivational qualities of music in exercise

    Music can enhance athletic performance in different ways, depending on whether it is synchronous (movements are in time with rhythm; Terry et al., 2006), asynchronous (no conscious synchronization between movement and music tempo; Terry et al., 2006), or pre-task (to regulate arousal level prior to performance; Karageorghis et al., 2012). Benefits of music include more positive mood, reduced pain/fatigue, reduced subjective exertion, improved performance (e.g. in treadmill running, cycling, on grip strength, etc.), faster acquisition of motor skills, and increased likelihood of flow states (Karageorghis et al., 2001). Motivational qualities of a piece of music depend on the response to rhythm, pitch/harmony, identification with cultural groups, and non-musical associations (Karageorghis, et al., 1999; Terry et al., 2011). Tempo is the most important determinant of response to music (Karageorghis et al., 1999) and an average of 130 bpm was reported to be most suitable for workout settings (Felstead et al, 2006) and for workouts with low to moderate intensity (Karageorghis et al., 2006). This mixed-methods project focuses on the motivational qualities of music, extending Priest’s findings (2003). Which music enhances motivation during a workout? Is music particularly designed for workouts more likely to motivate than other music? A total of 24 well-known sound samples with a tempo close to 130 bpm will be cut to a length of ten seconds and assigned to eight different musical styles, making them three samples for each style. Four styles will reflect workout- music (taken from Spotify workout playlists), and four will be non-workout music including Jazz, Classical, Pop/Rock, and Electronic music. Regular gym-goers will be asked to listen to each sample in a random order and to answer the question, how much they would like to listen to that music during a workout, using a questionnaire after each sample. Participants are requested to rate each sample on a seven-point-scale and briefly comment on it. In a second run, subjects will be asked about their musical preferences in general and to rate the samples’ musical styles on a seven-point-scale accordingly.
