    Cytokeratin positivity in paraffin-embedded malignant melanomas : comparative study of KL1, A4 and Lu5 antibodies

    The unclear role of cytokeratin (CK) in the progression and diagnostics of malignant melanomas stimulated us to compare the reactivity of three antibodies directed to CK in 109 paraffin-embedded melanomas. By far the majority of melanomas did not express cytokeratin even at the<1% level, only vimentin. In about 6% of melanomas it was possible to find CK expression ranging between 3 and 40% of melanoma cells. There was a correlation between CK expression and pTstage. Cytokeratin-expressing tumours were found in the more advanced pT-stages. The independent prognostic values of none of the three CK antibodies investigated could be shown

    Quantitation of Formoterol, Salbutamol, and Salbutamol-4′-O-Sulfate in Human Urine and Serum via UHPLC-MS/MS

    The adrenergic beta-2 agonists formoterol and salbutamol are used for the treatment of asthma and COPD but are also misused in sports competitions. Therefore, they are included in WADA regulations. Both drugs are mainly excreted in urine after administration via inhalation. A four-armed, double-blind cross-over clinical trial was conducted involving endurance-trained participants (12 females and 12 males). Inhalation dosages of 36 μg formoterol, 1200 μg salbutamol, a combination of both, or a placebo were administered before exercise. Serum and urine were collected after exercise and 3 and 24 h after administration. Here, we show the successful quantitation of formoterol, salbutamol, and its phase II metabolite salbutamol-4′-O-sulfate in all urine and serum samples using ultra-high performance liquid chromatography–tandem mass spectrometry. In the serum analysis, results of up to 14.2 pg/mL formoterol, 10.0 ng/mL salbutamol, and 21.4 ng/mL salbutamol-4′-O-sulfate (calculated as salbutamol equivalent) were found. In urine, maximum concentrations (after deglucuronidation) were 17.2 ng/mL formoterol, 948.5 ng/mL salbutamol, and 2738.5 ng/mL salbutamol-4′-O-sulfate. Sex-specific differences in serum concentrations as well as in urinary excretion were observed.     Systematic symptom screening in patients with advanced cancer treated in certified oncology centers: results of the prospective multicenter German KeSBa project

    Purpose Guidelines recommend a structured symptom screening (SC) for especially advanced cancer patients (CPs). The aim of this multicenter German prospective quality assurance project KeSBa (Kennzahl Symptom- und Belastungserfassung) was to gain knowledge on SC procedures in Oncology Centers (OCs) for advanced cancer patients and a first impression on the consequences of SC. Methods The KeSBa project consisted of three phases: pilot, 3 months screening and feedback phase. Participating OCs decided to use either the Minimal Documentation System (MIDOS) or the Integrated Palliative care Outcome Scale (IPOS) and defined the cutoff values for positive screening results. Results Out of 172 certified German OCs, 40 (23%) participated in the KeSBa pilot phase, 29 (16.8%) in the 3 months screening phase using MIDOS ( n  = 18, 58.6%) or IPOS ( n  = 11, 41.3%) and in the feedback round. 25/29 performed paper-based screening (86.2%). 2.963 CPs were screened. Results were documented for 1255 (42.2%, SC +) positive and 874 (29.5%, SC–) negative screenings depending on the center´s schedules: 452 SC + CPs (28.4%) and 42 SC– CPs (2.6%) had contact to specialized palliative care or other supportive specialist teams afterwards, 458 SC + CPs (28.8%) and 605 SC– CPs (38.1%) remained in standard oncology care. In the feedback round missing resources (personal and IT) and improved communication were mentioned most often. Conclusion Routine SC is feasible in advanced CPs treated in OCs but associated with considerable workload. In 42.2% of CPs SC was classified as positive, indicating the need of further diagnostics or professional judgment.     The Convective and Orographically-induced Precipitation Study (COPS): the scientific strategy, the field phase, and research highlights

    Within the framework of the international field campaign COPS (Convective and Orographically-induced Precipitation Study), a large suite of state-of-the-art meteorological instrumentation was operated, partially combined for the first time. This includes networks of in situ and remote-sensing systems such as the Global Positioning System as well as a synergy of multi-wavelength passive and active remote-sensing instruments such as advanced radar and lidar systems. The COPS field phase was performed from 01 June to 31 August 2007 in a low-mountain area in southwestern Germany/eastern France covering the Vosges mountains, the Rhine valley and the Black Forest mountains. The collected data set covers the entire evolution of convective precipitation events in complex terrain from their initiation, to their development and mature phase until their decay. Eighteen Intensive Observation Periods with 37 operation days and eight additional Special Observation Periods were performed, providing a comprehensive data set covering different forcing conditions. In this article, an overview of the COPS scientific strategy, the field phase, and its first accomplishments is given. Highlights of the campaign are illustrated with several measurement examples. It is demonstrated that COPS research provides new insight into key processes leading to convection initiation and to the modification of precipitation by orography, in the improvement of quantitative precipitation forecasting by the assimilation of new observations, and in the performance of ensembles of convection-permitting models in complex terrain

    Forced Migration and Transnational Family Arrangements - Eritrean and Syrian Refugees in Germany (TransFAR): Methodology Report

    Forced migration and its impact on family dynamics is an important topic with high policy relevance. However, this intersection has received little scientific attention because quantitative surveys on refugee populations are scarce and mainly focus on individuals. Families, their transnational arrangements and social integration into the host society remain insufficiently researched. The project "Forced Migration and Transnational Family Arrangements - Eritrean and Syrian Refugees in Germany" (TransFAR) aims at filling this gap by determining the structure and spatial dispersion of forced migrants’ families, their family lives, as well as the way in which these interact with social life and participation in the host country. The project takes a comparative cross-national perspective by covering two of the major countries of origin of refugees in Germany: Eritrea and Syria. These two countries serve as illustrative examples of the migra-tion/flight dynamics and integration processes established between Sub-Saharan Africa and the Middle East on the one hand and European refugee-receiving countries on the other. The TransFAR project is based on a quantitative survey that was carried out in summer 2020, targeting individuals with Eritrean or Syrian citizen-ship aged 18-45 years at arrival, who migrated to Germany between June 2013 and June 2019. The nationally representative sample was drawn from the Central Register of Foreigners in Germany. The realised sample includes 1,458 persons from Eritrea and Syria (50% women, 50% men).
