Cet article a pour ambition de montrer en quoi le montage, que l'on décrit souvent comme l'essence du langage cinématographique, est un dispositif de médiation de la vision et de l'action. Walter Benjamin, en élargissant les analyses de Eisenstein sur le « montage des attractions », fut un des premiers théoriciens à souligner que le choc visuel du montage obligeait le spectateur à réagir sur le plan politique et à adopter un comportement progressiste.The aim of this article is to show how montage, often described as the essence of cinematographic language, constitutes an apparatus for the mediation of vision and action. Molecular structure, mechanical behavior and failure mechanism of the C-terminal cross-link domain in type I collagen
Collagen is a key constituent in structural materials found in biology, including bone, tendon, skin and blood vessels. Here we report a first molecular level model of an entire overlap region of a C-terminal cross-linked type I collagen assembly and carry out a nanomechanical characterization based on large-scale molecular dynamics simulation in explicit water solvent. Our results show that the deformation mechanism and strength of the structure are greatly affected by the presence of the cross-link, and by the specific loading condition of how the stretching is applied. We find that the presence of a cross-link results in greater strength during deformation as complete intermolecular slip is prevented, and thereby particularly affects larger deformation levels. Conversely, the lack of a cross-link results in the onset of intermolecular sliding during deformation and as a result an overall weaker structure is obtained. Through a detailed analysis of the distribution of deformation by calculating the molecular strain we show that the location of largest strains does not occur around the covalent bonding region, but is found in regions further away from this location. The insight developed from understanding collagenous materials from a fundamental molecular level upwards could play a role in advancing our understanding of physiological and disease states of connective tissues, and also enable the development of new scaffolding material for applications in regenerative medicine and biologically inspired materials
LuxCDE-luxAB-based promoter reporter system to monitor the Yersinia enterocolitica O : 3 gene expression in vivo
It is crucial to understand the in vitro and in vivo regulation of the virulence factor genes of bacterial pathogens. In this study, we describe the construction of a versatile reporter system for Yersinia enterocolitica serotype O:3 (YeO3) based on the luxCDABE operon. In strain YeO3-luxCDE we integrated the luciferase substrate biosynthetic genes, luxCDE, into the genome of the bacterium so that the substrate is constitutively produced. The luxAB genes that encode the luciferase enzyme were cloned into a suicide vector to allow cloning of any promoter-containing fragment upstream the genes. When the obtained suicide-construct is mobilized into YeO3-luxCDE bacteria, it integrates into the recipient genome via homologous recombination between the cloned promoter fragment and the genomic promoter sequence and thereby generates a single-copy and stable promoter reporter. Lipopolysaccharide (LPS) O-antigen (O-ag) and outer core hexasaccharide (OC) of YeO3 are virulence factors necessary to colonization of the intestine and establishment of infection. To monitor the activities of the OC and O-ag gene cluster promoters we constructed the reporter strains YeO3-P-oc::luxAB and YeO3-P-op1::luxAB, respectively. In vitro, at 37 degrees C both promoter activities were highest during logarithmic growth and decreased when the bacteria entered stationary growth phase. At 22 degrees C the OC gene cluster promoter activity increased during the late logarithmic phase. Both promoters were more active in late stationary phase. To monitor the promoter activities in vivo, mice were infected intragastrically and the reporter activities monitored by the IVIS technology. The mouse experiments revealed that both LPS promoters were well expressed in vivo and could be detected by IVIS, mainly from the intestinal region of orally infected mice.Peer reviewe

Визначено можливості впровадження схем гарантування та мікрострахування підприємств малого бізнесу в Україні. Проанализированы основные параметры деятельности фондов гарантирования кредитов и институтов микрострахования. Glucose-6-Phosphatase Catalytic Subunit 3 (G6PC3) Deficiency Associated With Autoinflammatory Complications
G6PC3 deficiency typically causes severe congenital neutropenia, associated with susceptibility to infections, cardiac and urogenital abnormalities. However, here we describe two boys of Pakistani origin who were found to have G6PC3 deficiency due to c.130 C>T mutation, but who have clinical phenotypes that are typical for a systemic autoinflammatory syndrome. The index case presented with combination of unexplained fevers, severe mucosal ulcers, abdominal symptoms, and inflammatory arthritis. He eventually fully responded to anti-TNF therapy. In this study, we show that compared with healthy controls, neutrophils and monocytes from patients have reduced glycolytic reserve. Considering that healthy myeloid cells have been shown to switch their metabolic pathways to glycolysis in response to inflammatory cues, we studied what impact this might have on production of the inflammatory cytokines. We have demonstrated that patients’ monocytes, in response to lipopolysaccharide, show significantly increased production of IL-1β and IL-18, which is NLRP3 inflammasome dependent. Furthermore, additional whole blood assays have also shown an enhanced production of IL-6 and TNF from the patients’ cells. These cases provide further proof that autoinflammatory complications are also seen within the spectrum of primary immune deficiencies, and resulting from a wider dysregulation of the immune responses
Orthodontics in Relation with Alveolar Bone Grafting in CLP Patients
Alveolar bone grafting is an essential step in the overall management of patients with cleft lip and palate (CLP). The numerous advantages of this procedure have been reported in the literature. Failure to rehabilitate the alveolar cleft may give rise to a variety of problems. Lack of investing alveolar bone often precludes the correction of anterior tooth irregularities and limits orthodontic treatment and/or prosthodontic rehabilitation. The success of the graft is multifactorial. The periodontal health of the surrounding graft tissues, the experience and ability of the surgeon, the timing of surgery, and orthodontic management of the cleft area before and after grafting are shown to be the most important factors in this issue. In this chapter, current orthodontic approaches in relation with alveolar bone grafting (ABG) in cleft patients will be discussed
