Benchmarking recombinant Pichia pastoris for 3-hydroxypropionic acid production from glycerol
The use of the methylotrophic yeast Pichia pastoris (Komagataella phaffi) to produce heterologous proteins has been largely reported. However, investigations addressing the potential of this yeast to produce bulk chemicals are still scarce. In this study, we have studied the use of P. pastoris as a cell factory to produce the commodity chemical 3-hydroxypropionic acid (3-HP) from glycerol. 3-HP is a chemical platform which can be converted into acrylic acid and to other alternatives to petroleum-based products. To this end, the mcr gene from Chloroflexus aurantiacus was introduced into P. pastoris. This single modification allowed the production of 3-HP from glycerol through the malonyl-CoA pathway. Further enzyme and metabolic engineering modifications aimed at increasing cofactor and metabolic precursors availability allowed a 14-fold increase in the production of 3-HP compared to the initial strain. The best strain (PpHP6) was tested in a fed-batch culture, achieving a final concentration of 3-HP of 24.75 g l−1, a product yield of 0.13 g g−1 and a volumetric productivity of 0.54 g l−1 h−1, which, to our knowledge, is the highest volumetric productivity reported in yeast. Early-succession secondary forests following agropastoral abandonment are key winter habitats for the conservation of a priority bird in the European Alps
AbstractIn contrast to old-growth forests, early-successional stands remain understudied despite potentially harbouring species of conservation interest. With this work, focused on hazel grouse Tetrastes bonasia, a cryptic and indicator species known to select for close-to-natural forests, we evaluated winter densities, home range, microhabitat selection and diet, combining DNA-based mark-recapture and metabarcoding from faecal samples. In total, 216 droppings, collected over 2 years along forest transects in the Italian Alps, were successfully genotyped and 43 individuals were identified. Density estimates were similar to values reported by other studies in the Alps with an average of 4.5 and 2.4 individuals/km2 in the first and second study year, respectively, and mean home ranges estimated at 0.95 km2. According to habitat selection models and eDNA-based diet analysis, hazel grouse selected early-succession secondary-growth forests formed after the abandonment of traditional agropastoral activities. These forests, mostly composed of hazel Corylus avellana, Norway spruce Picea abies and Sorbus spp., provided winter food resources and shelter. The diet analysis also highlighted forest arthropods as a non-negligible source of food. Birds avoided areas subject to intensive browsing by ungulates; small forest roads seasonally closed to traffic had positive influence on hazel grouse (i.e. higher abundance of droppings), while roads open to traffic had no effect. Importantly, despite the high coverage of mature forest habitats of Community Interest (53% of our study area), droppings were more abundant in non-listed early-succession secondary forests with similar plant composition. Our results suggest that forest succession after agropastoral abandonment may be beneficial for some forest birds of conservation interest, while acknowledging its negative effects on the previous grassland biodiversity.

Posterior fossa tumours in childhood: Associated speech and language disorders post-surgery
Six children aged between 6 and 16 years who had undergone surgery for the removal of a posterior fossa tumour were assessed at least one year postoperatively to determine the incidence and severity of any associated speech or language deficits. Five males and one female were included in the sample. The subjects were administered a battery of speech/language assessments including: a language screening test, an articulation test, a dysarthria assessment and a perceptual speech analysis. The results indicated that dysarthria and/or language impairment occurs in some cases subsequent to surgical removal of posterior fossa tumours. The occurrence of muteness immediately post-surgery would appear to indicate a poor prognosis for speech abilities. A possible link between the occurrence of long term language disabilities in these children and post-surgical radiotherapy is documented

Histological and microbiological aspects of actinomycetoma cases in Venezuela
A ten year (1976-1986) review study of cases of Actinomycetoma in Venezuela was made through personal interview and clinical examinations, analysis of medical records of patients with actinomycetoma, histological studies of biopsy samples, as well as microbiological studies of isolates strain, also through out personal interviews with researchers and dermatologists who were sources of information on mycetoma cases. A total of 47 cases were recorded. As etiologic agent Actinomadura madurae was found in 20 cases - (42.5%), Nocardia brasiliensis in 13 cases (27.6%), Nocardia spp 7 cases (14.8%), Streptomyces somaliensis in 4 cases (8.5%), N. asteroides in 2 cases (4.2%) and N. otitidis caviarum, (N. caviae) in 1 case (2.1%). Most of the reported cases involved individuals living and working in rural areas, mostly males who outnumber females 4:1. The patients were 18 to 80 years old. A. madurae was reported as the most frequent etiologic agent. Most of the clinical cases were seen when the disease was well established. Twenty four of the forty seven cases reported were observed in Lara State, which represents a 51.0% of all the cases studied

Os autores descrevem episódios de mutismo após cirurgia para tumores da fossa posterior em pacientes jovens. São apresentados dois casos e é feita análise da ausência de expressão verbal que estabeleceu após a cirurgia. Também é feita análise do mutismo do ponto de vista de suas conotações neurológicas e psicológicas, ao mesmo tempo em que é revista a literatura sobre o assunto
