    Potential use of γδ T cell-based vaccines in cancer immunotherapy

    Immunotherapy is a fast advancing methodology involving one of two approaches: (1) compounds targeting immune checkpoints and (2) cellular immunomodulators. The latter approach is still largely experimental and features in vitro generated, live immune effector cells, or antigen-presenting cells. γδ T cells are known for their efficient in vitro tumor killing activities. Consequently, many laboratories worldwide are currently testing the tumor killing function of γδ T cells in clinical trials. Reported benefits are modest; however, these studies have demonstrated that large γδ T-cell infusions were well tolerated. Here, we discuss the potential of using human γδ T cells not as effector cells but as a novel cellular vaccine for treatment of cancer patients. Antigen-presenting γδ T cells do not require to home to tumor tissues but, instead, need to interact with endogenous, tumor-specific αβ T cells in secondary lymphoid tissues. Newly mobilized effector αβ T cells are then thought to overcome the immune blockade by creating proinflammatory conditions fit for effector T-cell homing to and killing of tumor cells. Immunotherapy may include tumor antigen-loaded γδ T cells alone or in combination with immune checkpoint inhibitors

    Pathological Effects of Aflatoxin and Their Amelioration by Vitamin E in White Leghorn Layers

    White Leghorn layer breeder hens, 30 weeks of age, were divided into 12 groups (A-L). Group A was kept on basal feed and served as control, while group B was offered feed supplemented with vitamin E (100 mg/Kg). Groups C-G were offered feed containing 100, 500, 2,500, 5,000 and 10,000 µg/Kg aflatoxin B1 (AFB1), respectively, whereas groups H-L were offered same dietary levels of AFB1 along with vitamin E (100 mg/Kg). The experimental feeds were offered for three weeks and afterward all the groups were switched over to basal feed for next two weeks. Body weight, absolute and relative weights of liver and kidneys of AF fed birds were significantly higher than control group. Pathological lesions in aflatoxin (AF) fed birds included enlarged, pale and friable liver, swollen kidneys and hemorrhages on different organs. Histopathological lesions in liver included fatty change, congestion and hemorrhages, while in kidneys tubular necrosis, cellular infiltration, congestion and hemorrhages were found in groups fed AFB1 at 500 μg/Kg and higher doses. In AF fed hens, no significant ameliorative effects of vitamin E could be observed upon AF induced decrease in feed intake, gross pathology and histopathological alterations and organ weight except body weights. It was concluded that the vitamin E ameliorated the AFB1 induced toxic effects in some of parameters studied

    Helping rural women in Pakistan to prevent postpartum hemorrhage: A quasi experimental study

    BACKGROUND: According to the Pakistan Demographic and Health Survey from 2006–2007, the maternal mortality ratio in rural areas is 319 per 100,000 live births. Postpartum hemorrhage is the leading cause of maternal deaths in Pakistan. The objectives of the study were to document the feasibility of distribution of misoprostol tablets by community-based providers mainly traditional birth attendants and acceptability and use of misoprostol by women who gave birth at home. METHODS: A quasi-experimental design, comprising intervention and comparison areas, was used to document the acceptability of providing misoprostol tablets to pregnant women to prevent postpartum hemorrhage in the rural community setting in Pakistan. Data were collected using structured questionnaires administered to women before and after delivery at home and their birth attendants. RESULTS: Out of 770 women who delivered at home, 678 (88%) ingested misoprostol tablets and 647 (84%) ingested the tablets after the birth of the neonate but prior to the delivery of the placenta. The remaining women took misoprostol tablets after delivery of the placenta. Side effects were experienced by 40% of women and were transitory in nature. Among women who delivered at home, 80% said that they would use misoprostol tablets in the future and 74% were willing to purchase them in the future. CONCLUSIONS: Self-administration of misoprostol in the home setting is feasible. Community-based providers, such as traditional birth attendants and community midwives with proper training and counseling, play an important role in reducing postpartum hemorrhage. Proper counseling and information exchange are helpful for introducing new practices in resource-constrained rural communities. Until such a time that skilled birth attendance is made more universally available in the rural setting, alternative strategies, such as training and using the services of traditional birth attendants to provide safe pregnancy care, must be considered

    Person Organization Fit, Organizational Commitment and Knowledge Sharing Attitude–An Analytical Study

    The purpose of this study is to analyze the relationship among Person Organization Fit (POF), Organizational Commitment (OC) and Knowledge Sharing Attitude (KSA). The paper develops a conceptual frame based on a theory and literature review. A quantitative approach has been used to measure the level of POF and OC as well as to explore the relationship of these variables with KSA & with each other by using a sample of 315 academic managers of public sector institutions of higher education. POF has a positive relationship with OC and KSA. A positive relationship also exists between OC and KSA. It would be an effective contribution in the existing body of knowledge. Managers and other stakeholders may be helped to recognize the significance of POF, OC and KSA as well as their relationship with each other for ensuring selection of employee’s best fitted in the organization and for creating and maintaining a conducive environment for improving organizational commitment and knowledge sharing of the employees which will ultimately result in enhanced efficacy and effectiveness of the organization

    ABSTRACT: This paper aims at finding the leak rate through ANSI class#150 flange joints using compressed asbestos sheet gasket under combined structural and thermal transient loading conditions. The solution is obtained using two different leak rate models and two different bolt up values. The gasket compressive strain based model employs strains that are determined using finite element analysis. The other model is based on the porous media theory in which gasket is considered as porous media. Leak rates determined using these leak rate models are compared for different tightness classes and discussed. ABSTRAK: Kajian bertujuan mencari kadar bocor menerusi sambungan bebibir kelas ANSI#150 menggunakan gasket kepingan asbestos termampat di bawah kondisi bebanan gabungan struktur dan terma fana. Solusinya diperolehi dengan menggunakan dua model kadar bocor yang berbeza dan dua nilai atas bolt yang berlainan. Model terikan berasaskan pemampat gasket menggunakan terikan yang ditentukan dengan analisis unsur terhingga.     Bullet-induced synovitis as a cause of secondary osteoarthritis of the hip joint: a case report and review of literature

    Abstract BACKGROUND: With increasing prevalence of gunshot injuries we are seeing more patients with retained bullet fragments lodged in their bodies. Embedded lead bullets are usually considered inert after their kinetic energy has dissipated hence these are not removed routinely. However, exposure of any foreign body to synovial fluid may lead to rapid degradation and hence result in systemic absorption, causing local and systemic symptoms. We present the case of a thirty year old man who came to our out patient department with a history of progressive, severe hip pain ten years after a gun shot injury to his right hip. CONCLUSION: The common belief that intraarticular bullets should not be removed has no benefit and may result in unwanted long term complications

    Devolved Ontology in Practice for a Seamless Semantic Alignment within Dynamic Collaboration Networks of SMEs

    The lack of a semantic alignment between collaborating small and medium enterprises causes frequent misinterpretations when exchanging information in the form of documents. If these companies are to achieve a seamless semantic alignment by exchanging documents, we should employ a conceptual model which does not rely on agreeing in advance on a centralised standard for document contents and format, but instead allows individual companies to maintain localised ontologies structuring their own documents allowing the companies to automatically establish a semantic alignment between pairs of collaborating companies, taking into account the ripple effects that such an alignment could trigger. In this article we demonstrate how the conceptual model of devolved ontology is engineered and tested to support such a scenario: we show how we have engineered the devolved ontology through a case study, and present experimental results on the document alignment intrinsically needed for this
