Las espigas de aparición tardía en trigo : condiciones que determinan su ocurrencia y consecuencias sobre el rendimiento
p.35-45Se evaluó la aparición tardía de espigas en el cultivo de trigo, para la sub región triguera II N argentina, discutiéndose las consecuencias sobre el rendimiento y sus componentes. A estos efectos se utilizaron resultados de ensayos multi locales (5 sitios) durante dos campañas (1982-1983) donde se varió la dosis y el momento de aplicación de nitrógeno. Se demostró la incidencia de la disponibilidad y el momento de aplicación del nitrógeno sobre la magnitud de este grupo de espigas, en condiciones de sequía en encañazón seguidas por importantes lluvias en pre floración, pudiendo estas representar el 40 por ciento de las espigas totales a cosecha. La contribución de estas espigas sobre el rendimiento se redujo a cerca de la mitad de la ejercida sobre el número de granos por metro cuadrado. Esto se debió a que los pesos de milgranos de este grupo de espigas fueron afectados por altas temperaturas durante su periodo de llenado, retrasando respecto al primer grupo. Se destaca el interés para el agrónomo de cuantificar este fenómeno para mejorar las estimaciones de rendimientos y los estudios de fechas de adición de nitrógeno
Reduced capacity of antibodies from patients infected with human immunodeficiency virus type 1 (HIV-1) group O to neutralize primary isolates of HIV-1 group M viruses
Neutralizing antibody patterns in sera of persons infected with human immunodeficiency virus type 1 (HIV-1) groups M and a to their homologous and heterologous primary isolates were determined in a peripheral blood mononuclear cell-based neutralization assay and correlated with their ability to bind to V3 loop synthetic peptides. Most HIV-1 group M sera (9/16) neutralized HIV-1 group a viruses, whereas fewer group a sera (3/13) only weakly neutralized HIV-1 group M viruses. Group M sera neutralizing HIV-1 group a viruses neutralized other HIV-1 group M viruses with titers of 1:10-1:1280. V3 loop binding capacity of sera did not reflect their neutralizing capacity of the homologous isolate. Despite the reduced neutralizing capacity of group a-infected patients ' sera to group M viruses, some group M- infected patients ' sera neutralized both HIV-1 group M and a isolates, suggesting that they share some conserved neutralizing epitopes. REVISITING ANNA MOSCOWITZ\u27S KROSS\u27S CRITIQUE OF NEW YORK CITY\u27S WOMEN\u27S COURT: THE CONTINUED PROBLEM OF SOLVING THE PROBLEM OF PROSTITUTION WITH SPECIALIZED CRIMINAL COURTS
This article explores New York City\u27s non-traditional, judicially based response to prostitution. This article first recounts the history of New York City’s Women’s Court. It then examines the work of the Midtown Community Court, the “problem-solving court” established in 1993 to address criminal issues, like prostitution, in Midtown Manhattan. It also discusses the renewed concerns about sex work in New York and describe the movement, propelled by modern reformers, to address prostitution through specialty courts. It then contrasts the shared features and attributes of the Women’s Court and Midtown Court models. Finally, the article urges modern reformers to step back from the problem-solving court movement and their call for the creation of more such specialized criminal courts

Van april 2006 tot en met juni 2007 voerde het VIOE (nu Onroerend Erfgoed) een preventief archeologisch onderzoek uit op het bedrijventerrein Menen-Oost-Uitbreiding. De opgravingen leverden vooral structuren en vondsten op van een kleine inheems-Romeinse nederzetting bestaande uit meerdere grachtensystemen, een aantal erven en een grafveld. Het betreft een meerfasige nederzetting die bewoond was van het midden van de 1ste eeuw tot het einde van de 3de eeuw na Chr. Het belang van de opgraving en de uitwerking van de site Menen-Kortewaagstraat ligt in het volledige beeld dat de site ophangt van een kleine inheems-Romeinse nederzetting in het zandlemige deel van het Menapische gebied. Omdat de site zowel een bewoningszone als een grafveld omvat, kon er informatie over heel wat facetten van het leven in de nederzetting worden ingewonnen, zoals de inrichting van de nederzetting, de aard van de bewoning, de economische activiteiten, rituele handelingen en de zorg voor de doden

Activity-based protein profiling has emerged as a powerful tool for visualizing glycosidases in complex biological samples. Several configurational cyclophellitol isomers have been shown to display high selectivity as probes for glycosidases processing substrates featuring the same configuration. Measurement of S-phase duration of adult stem cells in the flatworm Macrostomum lignano by double replication labelling and quantitative colocalization analysis
Platyhelminthes are highly attractive models for addressing fundamental aspects of stem cell biology in vivo. These organisms possess a unique stem cell system comprised of neoblasts that are the only proliferating cells during adulthood. We have investigated T-s (S-phase duration) of neoblasts during homoeostasis and regeneration in the flatworm, Macrostomum lignano. A double immunohistochemical technique was used, performing sequential pulses with the thymidine analogues CldU (chlorodeoxyuridine) and IdU (iododeoxyuridine), separated by variable chase times in the presence of colchicine. Owing to the localized nature of the fluorescent signals (cell nuclei) and variable levels of autofluorescence, standard intensity-based colocalization analyses could not be applied to accurately determine the colocalization. Therefore, an object-based colocalization approach was devised to score the relative number of double-positive cells. Using this approach, T-s (S-phase duration) in the main population of neoblasts was similar to 13 h. During early regeneration, no significant change in T-s was observed
A Specific Activity-Based Probe to Monitor Family GH59 Galactosylceramidase, the Enzyme Deficient in Krabbe Disease
Galactosylceramidase (GALC) is the lysosomal β-galactosidase responsible for the hydrolysis of galactosylceramide. Inherited deficiency in GALC causes Krabbe disease, a devastating neurological disorder characterized by accumulation of galactosylceramide and its deacylated counterpart, the toxic sphingoid base galactosylsphingosine (psychosine). We report the design and application of a fluorescently tagged activity-based probe (ABP) for the sensitive and specific labeling of active GALC molecules from various species. The probe consists of a β-galactopyranose-configured cyclophellitol-epoxide core, conferring specificity for GALC, equipped with a BODIPY fluorophore at C6 that allows visualization of active enzyme in cells and tissues. Detection of residual GALC in patient fibroblasts holds great promise for laboratory diagnosis of Krabbe disease. We further describe a procedure for in situ imaging of active GALC in murine brain by intra-cerebroventricular infusion of the ABP. In conclusion, this GALC-specific ABP should find broad applications in diagnosis, drug development, and evaluation of therapy for Krabbe disease
Evolution of antibody landscape and viral envelope escape in an HIV-1 CRF02_AG infected patient with 4E10-like antibodies
<p>Abstract</p> <p>Background</p> <p>A minority of HIV-1 infected individuals develop broad cross-neutralizing (BCN) plasma antibodies that are capable of neutralizing a spectrum of virus variants belonging to different HIV-1 clades. The aim of this study was to identify the targeted epitopes of an individual with BCN plasma antibodies, referred to as ITM4, using peptide phage display. This study also aimed to use the selected mimotopes as tools to unravel the evolution of the antibody landscape and the viral envelope escape which may provide us with new insights for vaccine design.</p> <p>Results</p> <p>This study led us to identify ITM4 plasma antibodies directed to the 4E10 epitope located in the gp41 membrane-proximal external region (MPER). Analysis of antibody specificities revealed unusual immunogenic properties of the ITM4 viral envelope, as not only the V3 loop and the gp41 MPER but also the C1 and lentivirus lytic peptide 2 (LLP2) region seem to be targets of the immune system. The 4E10-like antibodies are consistently elicited during the 6-year follow up period. HIV-1 ITM4 pseudoviruses showed an increasing resistance over time to MPER monoclonal antibodies 4E10 and 2F5, although no changes are found in the critical positions of the epitope. Neutralization of COT6.15 (subtype C; 4E10-sensitive) pseudoviruses with alanine substitutions in the MPER region indicated an overlapping specificity of the 4E10 monoclonal antibody and the ITM4 follow up plasma. Moreover the 4E10-like antibodies of ITM4 contribute to the BCN capacity of the plasma.</p> <p>Conclusions</p> <p>Using ITM4 BCN plasma and peptide phage display technology, we have identified a patient with 4E10-like BCN antibodies. Our results indicate that the elicited 4E10-like antibodies play a role in virus neutralization. The viral RNA was isolated at different time points and the ITM4 envelope sequence analysis of both early (4E10-sensitive) and late (4E10-resistant) viruses suggest that other regions in the envelope, outside the MPER region, contribute to the accessibility and sensitivity of the 4E10 epitope. Including ITM4 specific HIV-1 Env properties in vaccine strategies may be a promising approach.</p
