The definitive version was published in Journal of Geophysical Research: Oceans 121 (2016): 4077–4096, doi:10.1002/2015JC011177.Sea Surface Height (SSH) data from the European Centre for Medium-Range Weather Forecasts-Ocean Reanalysis System 4 (ECMWF-ORAS4) are used to determine the pressure difference in connection with variability of the South China Sea ThroughFlow (SCSTF) from 1958 to 2007. Two branches of SCSTF, the Karimata-Sunda Strait ThroughFlow (KSSTF) and the Mindoro Strait ThroughFlow (MSTF), are examined. Using the ensemble empirical mode decomposition method (EEMD), time series of pressure difference and volume transport are decomposed into intrinsic mode functions and trend functions, with the corresponding variability on different time scales. Pressure difference agrees with the KSSTF volume transport on decadal time scale; while for the MSTF, pressure difference varies similarly with volume transport on interannual time scale. Separating the dynamic height difference into the thermal and haline terms, for the KSSTF more than half of the dynamic height difference (32 cm) is due to the thermal contributions; while the remaining dynamic height difference (23 cm) is due to the haline contributions. For the MSTF, the dynamic height difference (29 cm) is primarily due to the thermal contribution (26 cm). This work was supported by the Strategic Priority Research Program of the Chinese Academy of Sciences (grant XDA11010304), the National Natural Science Foundation of China (grant number 41306015 and 41476013) and the Independent Research Project Program of State Key Laboratory of Tropical Oceanography (grant LTOZZ1603).2016-12-1

Simultaneous profiling of transcriptome and DNA methylome from a single cell.

BackgroundSingle-cell transcriptome and single-cell methylome technologies have become powerful tools to study RNA and DNA methylation profiles of single cells at a genome-wide scale. A major challenge has been to understand the direct correlation of DNA methylation and gene expression within single-cells. Due to large cell-to-cell variability and the lack of direct measurements of transcriptome and methylome of the same cell, the association is still unclear.ResultsHere, we describe a novel method (scMT-seq) that simultaneously profiles both DNA methylome and transcriptome from the same cell. In sensory neurons, we consistently identify transcriptome and methylome heterogeneity among single cells but the majority of the expression variance is not explained by proximal promoter methylation, with the exception of genes that do not contain CpG islands. By contrast, gene body methylation is positively associated with gene expression for only those genes that contain a CpG island promoter. Furthermore, using single nucleotide polymorphism patterns from our hybrid mouse model, we also find positive correlation of allelic gene body methylation with allelic expression.ConclusionsOur method can be used to detect transcriptome, methylome, and single nucleotide polymorphism information within single cells to dissect the mechanisms of epigenetic gene regulation

New Data on the Topside Electron Density Distribution

The existing uncertainties about the electron density profiles in the topside ionosphere, i.e., in the height region from hmF2 to approx. 2000 km, require the search for new data sources. The ISIS and Alouette topside sounder satellites from the sixties to the eighties recorded millions of ionograms and most were not analyzed in terms of electron density profiles. In recent years an effort started to digitize the analog recordings to prepare the ionograms for computerized analysis. As of November 2001 about 350,000 ionograms have been digitized from the original 7-track analog tapes. These data are available in binary and CDF format from the anonymous ftp site of the National Space Science Data Center. A search site and browse capabilities on CDAWeb assist the scientific usage of these data. All information and access links can be found at http://nssdc.gsfc.nasa.gov/space/isis/isis-status.html. This paper describes the ISIS data restoration effort and shows how the digital ionograms are automatically processed into electron density profiles from satellite orbit altitude (1400 km for ISIS-2) down to the F peak. Because of the large volume of data an automated processing algorithm is imperative. The automatic topside ionogram scaler with true height algorithm TOPIST software developed for this task is successfully scaling approx.70 % of the ionograms. An 'editing process' is available to manually scale the more difficult ionograms. The automated processing of the digitized ISIS ionograms is now underway, producing a much-needed database of topside electron density profiles for ionospheric modeling covering more than one solar cycle. The ISIS data restoration efforts are supported through NASA's Applied Systems and Information Research Program

This paper chooses closing price return rate series of EUR/USD to study. Sample interval covers from 22th July 2005 to 15th Sep 2008(before the financial crisis) and from 16th September 2008 to 19th May 2010(after the financial crisis). The author put forward semi-parameter estimation methods (Standard GPH Method, Tapered GPH Method), and concluded through comparable analysis that: In the conditions of V using T0.5、T0.525、T0.55、T0.575、T0.6 samples, standard GPH and tapered GPH tests are adopted. The results show that Fractal dimension parameter d is significantly greater than 0 and the statistics are more than critical value of 1% level before financial crisis both EUR/USD. After financial crisis, the parameter has become smaller than that before financial crisis, which is near 0 significantly. In the long term, there is no trend or structural breaks in the exchange market. This study's conclusion was that long-term memory exists in daily return time series of EUR/USD become smaller after financial crisis.
