    Population genetics of trypanosoma brucei rhodesiense: clonality and diversity within and between foci

    African trypanosomes are unusual among pathogenic protozoa in that they can undergo their complete morphological life cycle in the tsetse fly vector with mating as a non-obligatory part of this development. Trypanosoma brucei rhodesiense, which infects humans and livestock in East and Southern Africa, has classically been described as a host-range variant of the non-human infective Trypanosoma brucei that occurs as stable clonal lineages. We have examined T. b. rhodesiense populations from East (Uganda) and Southern (Malawi) Africa using a panel of microsatellite markers, incorporating both spatial and temporal analyses. Our data demonstrate that Ugandan T. b. rhodesiense existed as clonal populations, with a small number of highly related genotypes and substantial linkage disequilibrium between pairs of loci. However, these populations were not stable as the dominant genotypes changed and the genetic diversity also reduced over time. Thus these populations do not conform to one of the criteria for strict clonality, namely stability of predominant genotypes over time, and our results show that, in a period in the mid 1990s, the previously predominant genotypes were not detected but were replaced by a novel clonal population with limited genetic relationship to the original population present between 1970 and 1990. In contrast, the Malawi T. b. rhodesiense population demonstrated significantly greater diversity and evidence for frequent genetic exchange. Therefore, the population genetics of T. b. rhodesiense is more complex than previously described. This has important implications for the spread of the single copy T. b. rhodesiense gene that allows human infectivity, and therefore the epidemiology of the human disease, as well as suggesting that these parasites represent an important organism to study the influence of optional recombination upon population genetic dynamics

    A novel method to improve temperature simulations of general circulation models based on ensemble empirical mode decomposition and its application to multi-model ensembles

    A novel method based on the ensemble empirical mode decomposition (EEMD) method was developed to improve model performance. This method was evaluated by applying it to global surface air temperatures, which were simulated by eight general circulation models from the Coupled Model Intercomparison Project Phase 5 (CMIP5). The temperature simulations of the eight models were separated into their different components by EEMD. The model's performance improved after the first high-frequency component was removed from the original simulations by EEMD for each model, on both the global and continental scale. Moreover, EEMD was more effective in improving the model's performance compared to the wavelet transform method. The multi-model ensembles (MMEs) were calculated based on the EEMD-improved model simulations using the Average Ensemble Mean, Multiple Linear Regression, Singular Value Decomposition and Bayesian Model Averaging methods. The results showed that the MME forecasts performed better when the calculations were based on the EEMD-improved simulations as opposed to the original simulations on both the global and continental scale. Therefore, the results of the MME were further improved by using the EEMD-improved model simulations. This new method provides a simple way to improve model performance and can be easily applied to further improve MME simulations

    Do salivary bypass tubes lower the incidence of pharyngocutaneous fistula following total laryngectomy? A retrospective analysis of predictive factors using multivariate analysis

    Salivary bypass tubes (SBT) are increasingly used to prevent pharyngocutaneous fistula (PCF) following laryngectomy and pharyngolaryngectomy. There is minimal evidence as to their efficacy and literature is limited. The aim of the study was to determine if SBT prevent PCF. The study was a multicentre retrospective case control series (level of evidence 3b). Patients who underwent laryngectomy or pharyngolaryngectomy for cancer or following cancer treatment between 2011 and 2014 were included in the study. The primary outcome was development of a PCF. Other variables recorded were age, sex, prior radiotherapy or chemoradiotherapy, prior tracheostomy, type of procedure, concurrent neck dissection, use of flap reconstruction, use of prophylactic antibiotics, the suture material used for the anastomosis, tumour T stage, histological margins, day one post-operative haemoglobin and whether a salivary bypass tube was used. Univariate and multivariate analysis were performed. A total of 199 patients were included and 24 received salivary bypass tubes. Fistula rates were 8.3% in the SBT group (2/24) and 24.6% in the control group (43/175). This was not statistically significant on univariate (p value 0.115) or multivariate analysis (p value 0.076). In addition, no other co-variables were found to be significant. No group has proven a benefit of salivary bypass tubes on multivariate analysis. The study was limited by a small case group, variations in tube duration and subjects given a tube may have been identified as high risk of fistula. Further prospective studies are warranted prior to recommendation of salivary bypass tubes following laryngectomy

    THE INTEGRATION OF REMOTE SENSING AND GEOGRAPHIC INFORMATION SYSTEM (GIS) IN MANAGING URBAN ECOSYSTEMS

    Urban ecosystems face numerous challenges due to rapid urbanization and population growth. Effective management of these ecosystems is crucial to ensure their sustainability and the well-being of urban residents. Remote sensing (RS) and Geographic Information Systems (GIS) have emerged as valuable tools for understanding and managing urban ecosystems. The integration of remote sensing and GIS technologies facilitate the monitoring and assessment of urban biodiversity, aiding in the conservation and restoration of ecological habitats. With this mind, the objective of this study was to investigate the integration of remote sensing and GIS technologies for real-time monitoring and assessment of environmental parameters in urban ecosystems, and their role in supporting sustainable urban ecosystem conservation efforts. Landsat 8 Operational Land Imager (OLI) images were acquired between January 2nd and April 5th 2020 to assess and monitor the dynamics in urban ecosystems in Abidjan, Accra, and Lagos. The Normalized Difference Built-up index was used to detect areas covered with concrete structures and impervious surfaces, while the Normalized Difference Vegetation Index and Normalized Difference Water Index were used to detect areas covered with vegetation and water bodies, respectively. Results of the study show that Abidjan, Accra, and Lagos experienced increased built-up areas at the expense of other land uses such as forests. Remote Sensing and GIS technologies provide valuable insights into the spatial and temporal dynamics of urban environments, supporting evidence-based decision-making and sustainable urban planning and development

    The role of large-scale spatial patterns in the chaotic amplification of perturbations in a Lorenz’96 model

    The preparation of perturbed initial conditions to initialize an ensemble of numerical weather forecasts is a crucial task in current ensemble prediction systems (EPSs). Perturbations are added in the places where they are expected to grow faster, in order to provide an envelope of uncertainty along with the deterministic forecast. This work analyses the influence of large-scale spatial patterns on the growth of small perturbations. Therefore, we compare Lyapunov vector (LV) definitions, used in the initialization of state-of-the-art EPSs, with the so-called characteristic LVs. We test the dynamical behaviour of these LVs in the two-scale Lorenz’96 system. We find that the commonly used definitions of LVs include non-intrinsic and spurious effects due to their mutual orthogonality. We also find that the spatial locations where the small-scale perturbations are growing are ‘quantized’ by the large-scale pattern. This ‘quantization’ enhances the artificial disposition of the LVs, which is only avoided using the characteristic LVs, an unambiguous basis which may also be of great use in realistic models for assessing or initializing EPSs

    Population structure of the malaria vector Anopheles moucheti in the equatorial forest region of Africa

    <p>Abstract</p> <p>Background</p> <p><it>Anopheles moucheti </it>is a major malaria vector in forested areas of Africa. However, despite its important epidemiological role, it remains poorly known and insufficiently studied. Here, levels of genetic differentiation were estimated between different <it>A. moucheti </it>populations sampled throughout its distribution range in Central Africa.</p> <p>Methods</p> <p>Polymorphism at ten microsatellite markers was compared in mosquitoes sampled in Cameroon, the Democratic Republic of Congo and an island on Lake Victoria in Uganda. Microsatellite data were used to estimate genetic diversity within populations, their relative long-term effective population size, and the level of genetic differentiation between them.</p> <p>Results</p> <p>All specimens collected in Tsakalakuku (Democratic Republic of Congo) were identified as <it>A. m. bervoetsi </it>while other samples consisted of <it>A. m. moucheti</it>. Successful amplification was obtained at all microsatellite loci within all <it>A. m. moucheti </it>samples while only six loci amplified in <it>A. m. bervoetsi</it>. Allelic richness and heterozygosity were high for all populations except the island population of Uganda and <it>A. m. bervoetsi</it>. High levels of genetic differentiation were recorded between <it>A. m. bervoetsi </it>and each <it>A. m. moucheti </it>sample as well as between the island population of <it>A. m. moucheti </it>and mainland populations. Significant isolation by distance was evidenced between mainland populations.</p> <p>Conclusion</p> <p>High levels of genetic differentiation supports complete speciation of <it>A. m. bervoetsi </it>which should henceforth be recognized as a full species and named <it>A. bervoetsi</it>. Isolation by distance is the main force driving differentiation between mainland populations of <it>A. m. moucheti</it>. Genetically and geographically isolated populations exist on Lake Victoria islands, which might serve as relevant field sites for evaluation of innovative vector control strategies.</p

    South American Plasmodium falciparum after the Malaria Eradication Era: Clonal Population Expansion and Survival of the Fittest Hybrids

    Malaria has reemerged in many regions where once it was nearly eliminated. Yet the source of these parasites, the process of repopulation, their population structure, and dynamics are ill defined. Peru was one of malaria eradication's successes, where Plasmodium falciparum was nearly eliminated for two decades. It reemerged in the 1990s. In the new era of malaria elimination, Peruvian P. falciparum is a model of malaria reinvasion. We investigated its population structure and drug resistance profiles. We hypothesized that only populations adapted to local ecological niches could expand and repopulate and originated as vestigial populations or recent introductions. We investigated the genetic structure (using microsatellites) and drug resistant genotypes of 220 parasites collected from patients immediately after peak epidemic expansion (1999–2000) from seven sites across the country. The majority of parasites could be grouped into five clonal lineages by networks and AMOVA. The distribution of clonal lineages and their drug sensitivity profiles suggested geographic structure. In 2001, artesunate combination therapy was introduced in Peru. We tested 62 parasites collected in 2006–2007 for changes in genetic structure. Clonal lineages had recombined under selection for the fittest parasites. Our findings illustrate that local adaptations in the post-eradication era have contributed to clonal lineage expansion. Within the shifting confluence of drug policy and malaria incidence, populations continue to evolve through genetic outcrossing influenced by antimalarial selection pressure. Understanding the population substructure of P. falciparum has implications for vaccine, drug, and epidemiologic studies, including monitoring malaria during and after the elimination phase

    Responding to globalisation: The case of elite artistic gymnastics in China

The concept of globalisation and patterns of ‘reach’ and ‘response’ act as the heuristic devices underpinning the analysis. Data were collected from a range of documents and fromand six semi-structured interviews. The trajectory of Chinese elite artistic gymnastics’ responses to globalisation can be characterised as a passive response in the 1950s, a participative response in the first half of the 1960s, a conflictual response from 1966 to the early 1970s, a participative response from the early 1970s to the 2012 Olympic Games, and a passive response, once again, during the Rio Olympiad (post-London 2012). In this way, a nation’s responses to globalisation are seen as dynamic rather than rigid or static. The ever-expanding reach of the international federation-led global system fundamentally demarcates the activities of member states. However, any nation-state has a degree of autonomy when responding to globalisation and in deciding the detailed pathways whereby to pursue success within this system, although in extreme cases, a nation-state can choose to resist this globalising context, largely depending on a government’s attitude towards globalisation and the value the government attaches to it
