Constant depth microfluidic networks based on a generalised Murray’s law for Newtonian and power-law fluids

using a novel biomimetic rule, based on Murray’s law. Murray’s principle is extended to
consider the flow of power-law fluids in planar geometries (i.e. of constant depth rectangular
cross-section) typical of lab-on-a-chip applications. The proposed design offers the ability to
control precisely the shear-stress distributions and to predict the flow resistance along the network.
We use an in-house code to perform computational fluid dynamics simulations in order
to assess the extent of the validity of the proposed design for Newtonian, shear-thinning and
shear-thickening fluids under different flow conditions
Red blood cells (RBCs) visualisation in bifurcations and bends
Bifurcating networks are commonly found in nature. One example is the microvascular system, composed of blood vessels consecutively branching into daughter vessels, driving the blood into the capillaries, where the red blood cells (RBCs) are responsible for delivering O 2 and up taking cell waste and CO 2 . In this preliminary study, we explore a microfluidic bifurcating geometry inspired by such biological models, for investigating RBC partitioning as well as RBC-plasma separation favored by the consecutive bifurcating channels. A biomimetic design rule [1] based on Murray’s law [2] was used to set the channels’ dimensions along the network, which consists of consecutive bifurcating channels of reducing diameter. The ability to apply differential flow resistances by controlling the flow rates at the end of the network allowed us to monitor the formation of a cell-free layer (CFL) for different flow conditions at haematocrits of 1% and 5%. We have also compared the values of CFL thickness determined directly by the measurement on the projection image created from a stack of images or indirectly by analyzing the intensity profile in the same projection. The results obtained from this study confirm the potential to study RBC partitioning along bifurcating networks, which could be of particular interest for the separation of RBCs from plasma in point-of-care devices.JF would like to thank Professor Graça Minas and her coworkers for

Microfluidic converging/diverging channels optimised for homogeneous extensional deformation
In this work we optimise microfluidic converging/diverging geometries in order to produce constant strain-rates along the centreline of the flow, for performing studies under homogeneous extension. The design is examined for both two-dimensional and three-dimensional flows where the effects of aspect ratio and dimensionless contraction length are investigated. Initially, pressure driven flows of Newtonian fluids under creeping flow conditions are considered, which is a reasonable approximation in microfluidics, and the limits of the applicability of the design in terms of Reynolds numbers are investigated. The optimised geometry is then used for studying the flow of viscoelastic fluids and the practical limitations in terms of Weissenberg number are reported. Furthermore, the optimisation strategy is also applied for electro-osmotic driven flows, where the development of a plug-like velocity profile allows for a wider region of homogeneous extensional deformation in the flow field
Sulfated galactans from red seaweed Gracilaria fisheri target epidermal growth factor receptor (EGFR) and inhibit cholangiocarcinoma cells (CCA) proliferation
Cholangiocarcinoma (CCA) is increasing in incidence worldwide and is resistant to chemotherapeutic agents, making treatment of CCA a major challenge. Previous studies reported that natural sulfated polysaccharides (SPs) disrupted growth factor receptor activation in cancer cells. The present study, therefore, aimed at investigating the anti-proliferation effect of sulfated galactans (SG) isolated from the red seaweed Gracilaria fisheri (G. fisheri) on CCA cell lines. Direct binding activity of SG to CCA cells, epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) were determined. The effect of SG on proliferation of CCA cells was investigated. Cell cycle analyses and expression of signaling molecules associated with proliferation were also determined. The results demonstrated that SG bound directly to EGFR. SG inhibited proliferation of various CCA cell lines by inhibiting EGFR and extracellular signal-regulated kinases (ERK) phosphorylation, and inhibited EGF-induced increased cell proliferation. Cell cycle analyses showed that SG induced cell cycle arrest at the G0/G1 phase, down-regulated cell cycle genes and proteins (cyclin-D, cyclin-E, Cdk-4, Cdk-2), and up-regulated the tumor suppressor protein P53 and the cyclin-dependent kinase inhibitor P21. Taken together, these data demonstrate that SG from G. fisheri inhibited proliferation of CCA cells, and its mechanism of inhibition is mediated, to some extent, by inhibitory effects on EGFR activation and EGFR/ERK signaling pathway. SG presents a potential EGFR targeted molecule, which may be further clinically developed in a combination therapy for CCA treatment

univariate or multivariate family of distributions. An information theoretic
procedure is developed which is based on general divergence measures for
testing the hypothesis of the existence of a change. For comparing the accuracy
of the new test-statistic a simulation study is performed for the special case
of a univariate discrete model. Finally, the procedure proposed in this paper
is illustrated through a classical change-point example

Applications of stochastic modeling in air traffic management : Methods, challenges and opportunities for solving air traffic problems under uncertainty
In this paper we provide a wide-ranging review of the literature on stochastic modeling applications within aviation, with a particular focus on problems involving demand and capacity management and the mitigation of air traffic congestion. From an operations research perspective, the main techniques of interest include analytical queueing theory, stochastic optimal control, robust optimization and stochastic integer programming. Applications of these techniques include the prediction of operational delays at airports, pre-tactical control of aircraft departure times, dynamic control and allocation of scarce airport resources and various others. We provide a critical review of recent developments in the literature and identify promising research opportunities for stochastic modelers within air traffic management
Do institutional arrangements make a difference to transport policy and implementation? Lessons for Britain
This paper describes local government decision-making in transport in three areas of the UK, London, West Yorkshire and Edinburgh, in which major changes in local government decision-making structures have taken place over the last decade, and between which arrangements are now very different. The research discusses whether institutional change has had a beneficial or adverse effect, and whether any of the current structures provides a more effective framework for policy development and implementation. The results show that although the sites share a broadly common set of objectives there are differences in devolved responsibilities and in the extent to which various policy options are within the control of the bodies charged with transport policy delivery. The existence of several tiers of government, coupled with the many interactions required between these public sector bodies and the predominantly private sector public transport operators appears to create extra transactional barriers and impedes the implementation of the most effective measures for cutting congestion. There is, however, a compelling argument for the presence of an overarching tier of government to organise travel over a spatial scale compatible with that of major commuter patterns. The extent to which such arrangements currently appear to work is a function of the range of powers and the funding levels afforded to the co-ordinating organisation
An Optimisation Framework for Airline Fleet Maintenance Scheduling with Tail Assignment Considerations
Fierce competition between airlines has led to the need of minimising the operating costs while also ensuring quality of service. Given the large proportion of operating costs dedicated to aircraft maintenance, cooperation between airlines and their respective maintenance provider is paramount. In this research, we propose a framework to develop commercially viable and maintenance feasible flight and maintenance schedules. Such framework involves two multi-objective mixed integer linear programming (MMILP) formulations and an iterative algorithm. The first formulation, the airline fleet maintenance scheduling (AMS) with violations, minimises the number of maintenance regulation violations and the number of not airworthy aircraft; subject to limited workshop resources and current maintenance regulations on individual aircraft flying hours. The second formulation, the AMS with tail assignment (TA) allows aircraft to be assigned to different flights. In this case, subject to similar constraints as the first formulation, six lexicographically ordered objective functions are minimised. Namely, the number of violations, maximum resource level, number of tail reassignments, number of maintenance interventions, overall resource usage, and the amount of maintenance required by each aircraft at the end of the planning horizon. The iterative algorithm ensures fast computational times while providing good quality solutions. Additionally, by tracking aircraft and using precise flying hours between maintenance opportunities, we ensure that the aircraft are airworthy at all times. Computational tests on real flight schedules over a 30-day planning horizon show that even with multiple airlines and workshops (16000 flights, 529 aircraft, 8 maintenance workshops) our solution approach can construct near-optimal maintenance schedules within minutes
