    Obg proteins are a family of P-loop GTPases, conserved from bacteria to human. The Obg protein in Escherichia coli (ObgE) has been implicated in many diverse cellular functions, with proposed molecular roles in two global processes, ribosome assembly and stringent response. Here, using pre-steady state fast kinetics we demonstrate that ObgE is an anti-association factor, which prevents ribosomal subunit association and downstream steps in translation by binding to the 50S subunit. ObgE is a ribosome dependent GTPase; however, upon binding to guanosine tetraphosphate (ppGpp), the global regulator of stringent response, ObgE exhibits an enhanced interaction with the 50S subunit, resulting in increased equilibrium dissociation of the 70S ribosome into subunits. Furthermore, our cryo-electron microscopy (cryo-EM) structure of the 50S? ObgE? GMPPNP complex indicates that the evolutionarily conserved N-terminal domain (NTD) of ObgE is a tRNA structural mimic, with specific interactions with peptidyl-transferase center, displaying a marked resemblance to Class I release factors. These structural data might define ObgE as a specialized translation factor related to stress responses, and provide a framework towards future elucidation of functional interplay between ObgE and ribosome-associated (p) ppGpp regulators. Together with published data, our results suggest that ObgE might act as a checkpoint in final stages of the 50S subunit assembly under normal growth conditions. And more importantly, ObgE, as a (p) ppGpp effector, might also have a regulatory role in the production of the 50S subunit and its participation in translation under certain stressed conditions. Thus, our findings might have uncovered an under-recognized mechanism of translation control by environmental cues

    Non-oscillatory forward-in-time method for incompressible flows

    This research extends the capabilities of Non-oscillatory Forward-in-Time (NFT) solvers operating on unstructured meshes to allow for accurate simulation of incompressible turbulent flows. This is achieved by the development of Large Eddy Simulation (LES) and Detached Eddy Simulation (DES) turbulent flow methodologies and the development of parallel option of the flow solver. The effective use of LES and DES requires a development of a subgrid-scale model. Several subgrid-scale models are implemented and studied, and their efficacy is assessed. The NFT solvers employed in this work are based on the Multidimensional Positive Definite Advection Transport Algorithm (MPDATA) that facilitates novel implicit Large Eddy Simulation (ILES) approach to treating turbulence. The flexibility and robustness of the new NFT MPDATA solver are studied and successfully validated using well established benchmarks and concentrate on a flow past a sphere. The flow statistics from the solutions are compared against the existing experimental and numerical data and fully confirm the validity of the approach. The parallel implementation of the flow solver is also documented and verified showing a substantial speedup of computations. The proposed method lays foundations for further studies and developments, especially for exploring the potential of MPDATA in the context of ILES and associated treatments of boundary conditions at solid boundaries

    Transition metal single-atom supported on PC3 monolayer for highly efficient hydrogen evolution reaction by combined density functional theory and machine learning study

    It is essential to develop non-precious metal-based alternatives used in hydrogen evolution reaction (HER) due to high cost and scarcity of Pt-based catalysts. Herein, through density functional theory (DFT) calculations, the HER activity over 26 single-atom anchored phosphorus carbide (PC3) monolayer (TM@PC3) has been systematically investigated. Results indicate that ΔG*H of V, Fe, Nb, Mo, and Pd@PC3 are lower than that of Pt (1 1 1) catalyst, with 0.03, −0.03, −0.07, −0.04, and − 0.02 eV, respectively. By imposing the criterion window (−0.2 ≤ ΔG*H ≤ 0.2 eV), the d band centre (εd) for catalysts with excellent HER ability is in the range of − 0.68–0.41 eV. Besides, the five promising HER catalysts follow Volmer-Tafel mechanism. Fe, Nb, and Mo@PC3 show activation barriers of 0.75, 0.74, and 0.55 eV, lower than that of Pt. Machine learning (ML) was employed to explore the intrinsic relationship between catalytic performance and feature parameters. We demonstrated that the first ionization energy, bond length of TM − H and d band center are more correlated with hydrogen adsorption behaviour. Our work not only predicts that Fe, Nb, and Mo@PC3 can be substitutes for Pt metal in HER, but also reveals that the intrinsic correlation between catalytic activity and feature parameters by combining DFT and ML investigations.publishedVersio

    Study on the Evolution Mechanism of Lane Change Decision in Urban Expressway Diversion Area

    Urban expressway is the artery of modern urban traffic network, and the vehicle operation at off-ramp and diverging area affects the operation efficiency of the whole traffic system. The lane-changing game behavior between off ramp vehicles and going straight vehicles is very important in the whole driving behavior, and the lane-changing behavior between vehicles is easy to cause traffic accidents and stopping phenomena. To improve the driving efficiency in the diversion area and reduce the accident risk, by analyzing the process of the lane-changing behavior at off-ramp, this paper establishes a two-vehicle game model, solves the replicator dynamic equation according to the Dynamic Evolutionary Game Theory, uses MATLAB to calculate the evolution process and evolution speed based on different payoff factors, explores the influence of safety and speed on the stability of turn-out, and judges the evolutionary equilibrium point according to the determinant and trail of the Jacobi matrix. We build a realistic turn-out scenario and simulate it using the micro-traffic simulation software SUMO and it is found that: (1) The speed of different evolutionary equilibrium points based on speed payoff increased by 8.3% and 4.4% respectively compared with the speed of initial point. (2) The number of conflicts at the evolutionary equilibrium point based on the security payoff reduced to 22% of the initial point. (3) Compared with the initial point, the speed of the evolutionary stable point based on comprehensive payoff increased by 10.3%, and the number of conflicts reduced to 11% of the initial point. The simulation results show that the strategy of stable point of the evolutionary game model can effectively reduce the accident rate and improve the road operation efficiency

    Non-oscillatory forward-in-time integrators for viscous incompressible flows past a sphere

    A non-oscillatory forward-in-time (NFT) integrator is developed to provide solutions of the Navier-Stokes equations for incompressible flows. Simulations of flows past a sphere are chosen as a benchmark representative of a class of engineering flows past obstacles. The methodology is further extended to moderate Reynolds number, stably stratified flows under gravity, for Froude numbers that typify the characteristic regimes of natural flows past distinct isolated features of topography in weather and climate models. The key elements of the proposed method consist of the Multidimensional Positive Definite Advection Transport Algorithm (MPDATA) and a robust non-symmetric Krylov-subspace elliptic solver. The solutions employ a finite volume spatial discretisation on unstructured and hybrid meshes and benefit from a collocated arrangement of all flow variables while being inherently stable. The development includes the implementation of viscous terms with the detachededdy simulation (DES) approach employed for turbulent flows. Results demonstrate that the proposed methodology enables direct comparisons of the numerical solutions with corresponding laboratory studies of viscous and stratified flows while illustrating accuracy, robustness and flexibility of the NFT schemes.     Focusing on the clinical diagnosis and risk of hairy polyp: A report of 7 cases

    The hairy polyp is a rare congenital tumor. We introspected 7 cases of hairy polyp that were operated on in our hospital. Microscopically, they all contained only ectoderm and mesoderm tissue and the surface of tumors covered skin and hair regularly. One patient died of a hemorrhage and one girl`s hairy polyp occurred in nose. The result indicates that age, pedicle and thin hair are significant markers for the differential diagnosis of hairy polyp and it is important to closely follow a patient post operatively not only for airway obstruction but also hemorrhage

    The NS1 protein of influenza a virus interacts with heat shock protein Hsp90 in human alveolar basal epithelial cells: Implication for virus-induced apoptosis

    <p>Abstract</p> <p>Background</p> <p>Our previous study showed that the NS1 protein of highly pathogenic avian influenza A virus H5N1 induced caspase-dependent apoptosis in human alveolar basal epithelial cells (A549), supporting its function as a proapoptotic factor during viral infection, but the mechanism is still unknown.</p> <p>Results</p> <p>To characterize the mechanism of NS1-induced apoptosis, we used a two-hybrid system to isolate the potential NS1-interacting partners in A549 cells. We found that heat shock protein 90 (Hsp90) was able to interact with the NS1 proteins derived from both H5N1 and H3N2 viruses, which was verified by co-immunoprecitation assays. Significantly, the NS1 expression in the A549 cells dramatically weakened the interaction between Apaf-1 and Hsp90 but enhanced its interaction with cytochrome c (Cyt c), suggesting that the competitive binding of NS1 to Hsp90 might promote the Apaf-1 to associate with Cyt c and thus facilitate the activation of caspase 9 and caspase 3.</p> <p>Conclusions</p> <p>The present results demonstrate that NS1 protein of Influenza A Virus interacts with heat hock protein Hsp90 and meidates the apoptosis induced by influenza A virus through the caspase cascade.</p

    Comparative Pharmacokinetics and Preliminary Pharmacodynamics Evaluation of Piscidin 1 Against PRV and PEDV in Rats

    Antimicrobial peptide (Piscidin-1) is an effective natural polypeptide, which has great influence and potential on porcine epidemic diarrhea virus (PEDV) and pseudorabies virus (PRV). As an alternative antibiotic substitute, Piscidin-1 was subjected for pharmacokinetics study with three administration routes (i.v, i.m, and p.o) after a single dose of 2 mg/kg in rats and preliminary pharmacodynamics including antiviral activity in cell against PEDV and PRV. Based on 50 percent tissue culture infective dose (TCID50), there were about 2 and 10% virus survived ratios for Piscidin-1 against PRV and PEDV, respectively. The plaque test showed 1 and 2 μg/ml Piscidin-1 could eliminate 95% PRV and 85% PEDV, respectively. The main pharmacokinetics parameters of Cmax, AUC0−∞, Ke, t1/2, Tmax, MRT, and Clb in plasma were not applicable value, 25.9 μg*h/ml, 0.041 h−1, 16.97 h, not available value, 22.77 h, 0.067 L/h*kg after i.v administration, 2.37 μg/ml, 18.95 μg*h/ml, 0.029 h−1, 23.50 h, 0.33 h, 30.12 h, 0.095 L/h*kg after i.m administration and 0.73 μg/ml, 9.63 μg*h/ml, 0.036 h−1, 19.46 h, 0.50 h, 26.76 h, 0.171 L/h*kg after p.o administration. The bioavailability values after i.m and p.o administrations were calculated as 73.17 and 37.18%, respectively. The i.m administration was selected for pharmacokinetics study in ileum content against PEDV. The main pharmacokinetic parameters of Cmax, AUC0−∞, Ke, t1/2, Tmax, MRT, and Clb in ileum content were 1.67 μg/ml, 78.40 μg*h/ml, 0.034 h−1, 20.16 h, 8.12 h, 36.45 h, 0.026 L/h*kg. The Cmax values in plasma (2.37 μg/ml) and ileum content (1.67 μg/ml) were higher than the effective inhibitory concentration determined in the plaque test, and this indicates that Piscidin-1 might have effective inhibition effect against PRV and PEDV after administration of 2 mg/kg i.m. The results of this study represent the first investigations toward the pharmacokinetic characteristics of piscidin-1 in plasma upon three different administration routes, among which i.m. resulted in the highest bioavailability (73.17%). Furthermore, the pharmacokinetics study of ileum content indicated Piscidin-1 might have good effect against PEDV and could be regarded as an alternative antibiotic in clinical veterinary in the future study
